摘要
目的探讨抗氧化剂褪黑素(MEL)对间歇低氧(IH)小鼠胰岛素抵抗的干预作用及其可能机制。方法雄性C57BL/6J小鼠45只随机分成常氧对照组(NC组)、低氧对照组(CIH组)及低氧干预组(CIH+M组),每组各15只。成功复制动物模型后,测定各组小鼠胰岛素抵抗水平、胰腺组织中各种氧化应激指标。结果与NC组比较,CIH组胰岛素抵抗水平(30、60和90 min时间点)及胰腺组织中氧化应激水平显著增加(P<0.01);而CIH+M组上述各种指标较CIH组显著降低(P<0.01);CIH+M组与NC组比较上述各种指标无明显差异(P>0.05)。结论抗氧化剂MEL可通过其抗氧化作用干预IH所致胰岛素抵抗的发生。
[Objective] To explore the effect and mechanism of antioxidants melatonin (MEL) against in-termittent hypoxia (IH)-induced insulin resistance in mice. [Methods] Forty-five male C57BL/6J mouse wererandomly assigned to 3 groups: normoxic control group (NC group), conventional intermittent hypoxia group(CIH group), intermittent hypoxia MEL treatment group (CIH+M group), with 15 mouse in each group. Afterthe experiment, insulin resistance levels, various indexes of oxidative stress in pancreatic tissue were measuredin all groups. [Results] Compared with the NC group, the insulin resistance levels (30, 60 and 90 rain) andindexes of oxidative stress in pancreatic tissue were increased (all P〈0.01); But all above indexes were sig-nificantly lower in CIH+M group compared with those in CIH group (P 〈0.01); there were not significantlydifferent between CIH+M group and NC group in all above indexes (P〉0.05). [Conclusion] Antioxidant MELcould prevent CIH-induced insulin resistance by its anti-oxidative effect.
