摘要
以酶解液多肽指数为指标,选用胃蛋白酶、碱性蛋白酶、木瓜蛋白酶以及蜗牛酶、纤维素酶水解蛹虫草HWM剩余物,筛选蛹虫草HWM剩余物蛋白酶解最佳用酶。采用二次回归正交试验确定其最佳酶解工艺。试验结果表明,碱性蛋白酶为蛹虫草HWM剩余物蛋白降解的最佳用酶,其最佳酶解工艺参数为反应时间12 h,加酶量为干基底物质量的10%,底物质量浓度70 mg/mL,酶解温度55℃,初始pH 12.0。在此条件下,蛹虫草酶解液多肽指数达90.32%。
Pepsin, papain, alcalase, snailase and cellulase were selected to hydrolyze the protein residua of Cordyceps militaris to sift the optimum protease, TCA-SN% exponent was used as the index; the quadratic regression Orthogonal experiment was used to investigate the influence of factors on the TCA-SN% of Cordyceps militaris such as reaction time, substrate concentration, enzyme dosage, reaction temperature and initial pH. The results showed that the best pmtease is alcalase, and the optimum conditions were as follows: time 12 h, enzyme dosage 10%, concentration of substrate 70 mg/mL, reaction temperature 55 ℃ and pH 12.0 respectively, and under these conditions, the TCA-SN% reached 90.53%.
