摘要
以黑曲霉3.2130为出发菌株,采用紫外线-硫酸二乙酯复合诱变处理,得到一株酸性蛋白酶缺陷菌株B4,其酸性蛋白酶活性是原菌株的8.27%,并通过正交试验得出其最佳诱变条件:DES浓度1.5%,处理时间30 min,诱变温度40℃。对B4进行传代培养表明,该菌株具有良好的遗传稳定性,为异源β-葡萄糖苷酶基因在黑曲霉中的表达提供了先决条件。
Using Aspergillus niger 3.2130 as original strain, the deficient acidic protease producing strains were screened after compound mutagenesis of UV and diethy sulfate (DES), which named B4. The acidic protease activity was 8.27% of original strain. The optimum mutation parameters were as follows: 1.5% DES, and quivering for 30 min at 40 ℃. Genetic stability test showed that its acidic protease relative activity was stabilized, which provides the prerequisites for the heterogeneous beta-gtycosidase gene expression in the Aspergillus niger.
