Protective effects of N-acetylcysteine on hearing loss induced by short term noise exposure in guinea pigs

摘要 目的:研究N-乙酰半胱氨酸(NAC)对噪声性听力损伤的保护作用。方法:32只雄性白色豚鼠随机分成4组,每组8只。从噪声暴露前7天(7d)开始,分别灌胃给予蒸馏水、NAC 70、NAC 100、NAC 140mg/kg,1次/d,连续14d。在用药第8天(d 8),各组动物在中心频率4kHz、强度115dB(A)的倍频程噪声暴露4h,建立暴噪模型。测定噪声暴露前、噪声暴露后d 3、d 8、d 11各组豚鼠脑干听觉反应(ABR)的阈值。以听阈偏移反映听力受损情况,组织学检查耳蜗内外毛细胞的损伤程度,来评价NAC对豚鼠噪声性听力损伤的保护作用。结果:灌胃给予NAC 100、NAC 140mg/kg可在一定程度上减少噪声暴露后d 3、d 8、d 11豚鼠ABR的阈移值,减轻毛细胞的损伤。结论:NAC对噪声引起的急性和亚急性听力损伤均具有一定的保护作用。 Objective: To study the protective effect of N-acetylcysteine(NAC)on noise induced hearing loss (NIHL). Methods: Thirty-two male white guinea pigs were randomly divided into 4 groups, each consisting of 8 animals. Seven days before noise exposure,guinea pigs were administered distilled water (control), NAC 70, NAC 100 and NAC 1.40 mg/kg by gavage once daily for 14 days. On the 8th day(d 8), animals of the 4 groups were exposed to the noise of 4 kHz center frequency, 115 dB sound pressure level (SPL) for 4 hours to develop noise exposure model. The thresholds of auditory brainstem response (ABR) before noise exposure and 3, 8 and 11 days after the exposure were determined in the guinea pigs of the 4 groups. The damage extent of inner and outer hair cells of cochlea was evaluated histologically by scanning electron micro- scope. The protective effects of NAC on NIHL were evaluated by comparing the threshold shifts of A13R and the damage extent of bair cells between the ,1 groups. Results: The threshold shifts of ABR at days 3, 8 and 11 after noise exposure and damage of hair cells induced hy noise exposure could be decreased to some extent with the treatment of NAC 100, NAC 140 mg/kg. Conclusion: NAC has some protective effects on noise induced acute and sub-acute hearing loss.
