承德产道地药材黄芩不同采收期的根中黄芩苷的含量测定 被引量:11

Determination of Content of Baicalin from Scutellaria baicalensis roots of Chengde at Different Collecting Time

摘要 目的测定并比较河北省承德隆化县黄芩GAP种植基地所产的黄芩不同采收期的根中黄芩苷的含量。方法采用HPLC,色谱柱为AgilentZorbaxSB—C18(4.6mm×250mm,5μm),流动相为甲醇-水-甲酸(53:47:0.1),流速为0.8ml·min^-1,检测波长为280nm,柱温25℃进样量为10μl。结果黄芩苷在0.24—0.72μg内呈良好线性关系(r=0.9999),平均加样回收率98.14%,RSD2.00%(11,=6)。经比较发现,此批采的承德产黄芩的根中黄芩苷含量在6~10月总体呈下降趋势,2012年6月10目的黄芩的根中黄芩苷含量最高,达到21.25%,每个月份的第10日黄芩苷含量会变高。结论该方法简便、稳定可靠,重现性好,不同采收期的黄芩中黄芩苷的含量存在差异并有消长规律。 Objective To compare the content of Baicalin from which was produced at Hebei Provincial GAP planting area of Scutellaria baicalensis (Longhua country) at different collecting time. Methods Diosgenin was separated on a Agilont Zorbax SB - C18 (4.6 mm ×250 mm,5 μm) column and detected at 280nm at the flow rate of 0.8 ml ·min-1 using a mixture of methanol -H2O - formic acid (53: 47:0.1 ). The column temperature was set at room temperature and injection volume was 10μl. Re- sults The standard curve was linear in the range of 0.24 ~ 0.72 μg with correlation coefficient 0.9999. The average recovery of baicalin was 98.14% with RSD 2.0% ( n = 6 ). The total content of baicalin from Scutellaria baiealensis roots of Chengde from June to October decreased continuously,and the maxium value was June 10,2012 ,up to 21.25% ,the content of baicalin would be- come high in the tenth day of each month. Conclusion The method was simple, stable and reliable, reproducible, there were differences in the contents of baicalin and changed regularly.
