摘要
目的通过观察大鼠脑组织中水通道蛋白AQP-4的表达变化,探讨水通道蛋白4表达的变化与高湿环境的关系。方法 32只健康雄性的SD大鼠,随机分为30℃20d组、30℃30d组和30℃40d组(采用人工气候箱制作高湿模型)和常温常湿对照组,每组8只。采用实时定量PCR测定大鼠脑组织AQP-4 mRNA表达,Westem印迹法观察大鼠脑组织AQP-4蛋白表达。结果高湿环境下30℃20d组、30℃30d组和30℃40d组脑组织AQP-4 mRNA、蛋白水平与对照组相比下降,而只有30℃40d组脑组织AQP-4 mRNA、蛋白水平与对照组相比下降差异有统计学意义(P<0.05)。结论脑组织AQP-4表达的变化与高湿环境有关。
Objective To study the changes of expression of AQP-4 in the brain of rat in high humid environment. Meth-ods 32 healthy male SD rat were evenly randomized into 4groups:30℃ 20 day group, 30℃ 30 day group, 30℃ 40 day group and control group. Rats in high humid environment groups were exposed in an artificial environment chamber with ( RH) 〉90%. Real-time PCR and Western blotting assay were used to observe the expression changes of AQP-4 mRNA and protein in the brain of all the animals, respectively. Results The expression of AQP-4 mRNA and protein in the brain of 30℃ 40 day group signifi-cantly decreased compared to that of the control group. Conclusion The high humid environment has an effect on the changes of expression of AQP-4 in the brain.
