摘要
目的:观察丁基苯酞对单次全脑照射后大鼠大脑皮质水通道蛋白4(AQP4)表达的影响。方法:将雄性SD大鼠60只随机分为假照射组、单纯照射组和丁基苯酞组,采用10Gy单次全脑照射模型,制模后丁基苯酞组大鼠分别按剂量0.3、1.0和3.0mg/kg丁基苯酞腹腔注射;依文思蓝(EB)法评价血脑屏障通透性改变;免疫组织化学法检测皮质AQP4表达的变化。结果:单纯照射组大鼠脑组织EB含量为(1.62±0.18)μg/g,与假照射组的(0.58±0.11)μg/g比较,差异有统计学意义,P=0.006;照射后1周单纯照射组大鼠脑组织皮质AQP4表达(1.67±0.63)较对照组(4.00±0.32)降低,P=0.004。与单纯照射组比较,不同剂量丁基苯酞组大鼠脑组织EB含量均降低(0.3mg/kg组,P=0.002;1.0mg/kg组,P=0.004;3.0mg/kg组,P=0.003);照射后1周不同剂量丁基苯酞组皮质AQP4表达有不同程度增加,呈现剂量依赖性(0.3mg/kg组,P=0.035;1.0mg/kg组,P=0.042;3.0mg/kg组,P=0.006)。结论:大鼠全脑照射后皮质AQP4表达呈时程性变化,丁基苯酞对AQP4有调节作用。
OBJECTIVE:To observe the effect of DL-3-n-Butylphthalide on aquaporin-4(AQP4) expression in the cortex of modeling rats by whole brain irradiation.METHODS:Sixty male Sprague-Dawley rats were randomly divided into sham-irradiation group,irradiation group and DL-3-n-Butylphthalide group.The model of whole-brain irradiation was established with the exposure of the brain of the rats to 4Mev x-ray with a single-dose 10 Gy.The rats were injected intraperitoneally with DL-3-n-Butylphthalide at 0.3 mg/kg,1.0 mg/kg,3.0 mg/kg once a day.The permeability of blood brain barrier was measured by the amount of Evans blue(EB) in the brain and AQP4 expression in the cortex was examined by immunohistochemistry.RESULTS: Compared with sham-irradiation group(0.58±0.11) μg/g,the EB content in the brain tissue of irradiation group(1.62±0.18) μg/g significantly increased(P=0.006).The AQP4 expression in irradiation group was 1.67±0.63,which was decreased quickly compared with sham-irradiation group(4.00±0.32,P=0.004).Compared with irradiation group,The EB content in the brain tissue decreased significantly at different doses of DL-3-n-Butylphthalide groups(0.3 mg/kg group,P=0.002;1.0 mg/kg group,P=0.004;3.0 mg/kg group,P=0.003).And the AQP4 expression increased at 1 week post-radiation by a dose-dependent manner(0.3 mg/kg group,P=0.035;1.0 mg/kg group,P=0.042;3.0 mg/kg group,P=0.006).CONCLUSIONS: AQP4 expression in the cortex changed at a time-dependent manner after whole brain irradiation.DL-3-n-Butylphthalide could regulate the AQP4 expression in the cortex of modeling rats by whole brain irradiation.
