摘要
目的研究早产流产与胎盘细胞凋亡关系及其可能机制。方法随机选取正常妊娠足月分娩胎盘30例,早产胎盘30例,流产胎盘30例,取胎盘组织制备单细胞悬液,FCM检测胎盘细胞的凋亡率;DNA琼脂糖电泳检测胎盘细胞的凋亡特征性DNA条带;westernblot检测细胞凋亡相关蛋白表达。结果早产和流产胎盘细胞的凋亡率,明显高于正常妊娠分娩的胎盘细胞的凋亡率(P<0.05);同时琼脂糖凝胶电泳出现典型的凋亡细胞DNA梯形条带,伴随着bcl-2蛋白表达下调,bax、cyt-c、caspase-9和caspase-3蛋白表达上调,bcl-2/bax比值下降。结论早产流产可能与胎盘细胞启动线粒体途径凋亡密切相关。
Objective The aim of this study is to investigate apoptosis of placental cells via mitochondrial way and its the relationship between premature delivery/abortion and possible molecular mechanism. Methods 30 placenta of term delivery ,30 placenta of premature delivery, and 30 placenta of abortion were selected randomly. The suspension of single placental cells was made, and apoptosis rate of placental cells was detected by FCM, the ladder - shaped band was detected by DNA agarose gel electrophoresis, the expression of apoptosis related proteins was assessed by western blot. Results There was significant difference in apoptosis of placental cells between premature delivery/abortion, compared with term delivery group (P 〈 0.05 ). DNA agarose gel electrophoresis showed the typical ladder - shaped band of apoptosis, accompanied by the down - regulation of Bcl - 2 protein, and the up - regulation of Bax, Cyt - c, Caspase - 9 and Caspase - 3 protein, and the ratio of Bcl - 2/Bax decreased. Conclusion Premature delivery/abortion may be related to triggering placental cells apoptosis via the mitochondrial pathway.
