摘要
为提高卑霉素发酵体系的产量,对其操作条件和培养基进行优化。通过响应曲面法设计,得出最佳接种量为6%,摇床转速200r/min,在28℃发酵培养40h。得到优化的培养基组合为:豆饼粉4.36%,甘露醇1.8%,葡萄糖0.98%,可溶性淀粉0.24%,硫酸铵0.36%,碳酸钙0.5%,硫酸锌0.009 2%,硫酸镁0.013 1%,磷酸二氢钾0.000 5%,硫酸亚铁0.002 0%。研究结果表明,优化培养基比对照培养基的效价提高26.62%。
The operation condition and culture medium of avilamycin fermentation system are opti- mized for increase of production. Using response surface method, the optimized inoculation amount is 6%, and fermentation time is 40 h at 28 ℃ under 200 r/min shaking speed. The optimized medium combination is soybean powder 4.36% ,mannitol 1.8%,glucose 0.98%,soluble starch 0.24% ,ammo nium sulphate 0. 36%, calcium carbonate 0. 5 0. 013 1 % ,potassium dihydrogen phosphate 0.00 ue was increased by 26.62%. zinc sulphate 0. 009 2%, magnesium sulphate 0 5% ,ferrous sulphate 0. 002 0 %. The effective value was increased by 26.62%.
