摘要
目的:建立家兔血浆中阿西美辛和吲哚美辛浓度的测定方法。方法:采用高效液相色谱法,以酮洛芬为内标,色谱柱:Hypersil BDS C18(4.6 mm×250mm,5μm),流动相为pH 4.5醋酸盐缓冲液-乙腈-甲醇(50∶40∶10),流速为0.6 mL/min,柱温为25℃,检测波长为250nm,进样量为20μL。结果:阿西美辛在0.04-2.56μg/mL浓度范围内线性关系良好,平均回收率为89.4%,RSD=5.2%(n=5);吲哚美辛在0.03-1.92μg/mL浓度范围内线性关系良好,平均回收率为89.7%,RSD=5.7%(n=5)。结论:本法简单、灵敏、结果准确,可用于测定兔血浆中阿西美辛和吲哚美辛的浓度。
Objective:To establish a method for the determination of acemetacin and indometacin con-centrations in rabbits plasma .Method: HPLC method was adopted .Ketoprofen was used as the internal standard,Hypersil BDS C18 (4.6 mm×250mm,5μm) column was used,the mobile phase consisted pH4.5 acetate buffer solution-acetonitrile-methanol(50 ∶40 ∶10),flow rate was 0.6mL/min,column temperature was 25℃,the detection wave length was 250nm,injection volume was 20μL.Result: The linear range of acemetacin was 0.04-2.56μg/mL,the average recovery was 89.4%,RSD=5.2%(n=5);The linear range of indometacin was 0.03-1.92μg/mL,the average recovery was 89.7%,RSD=5.7%(n=5).Conclusion:The method is simple ,sensitive and accurate , and it can be applied to the determination of acemetacin and indo-metacin concentrations in rabbits plasma .
