摘要
目的通过对脓毒症大鼠血清中MMP-9浓度的影响并结合肺脏形态学及超微结构的变化,探讨血必净注射液对肺脏的保护作用及其作用机制。方法选用雄性S-D大鼠45只随机分为3组:空白组(Sham组)、盐水组(NS组)、血必净组(X组),每组15只。检测血清MMP-9水平、肺湿/干重比(W/D)、HE染色法光镜观察肺脏形态学改变,透射电镜观察肺脏血管内皮细胞的超微结构。结果与Sham组比较,NS组、X组大鼠肺湿/干重比均明显增大(P<0.01);与NS组比较,X组肺组织湿/干重比显著减小(P<0.01)。NS组、X组血清中MMP-9水平均显著高于Sham组(P<0.01);与NS组比较,X组血清中MMP-9水平下降(P<0.01)。Sham组大鼠肺组织结构及血管内皮细胞结构基本正常;NS组有明显炎性反应及血管内皮细胞结构破坏等;X组改变明显减轻但较Sham组重。结论血必净注射液可减少血清MMP-9水平,从而减轻肺血管内皮细胞损伤,改善肺血管通透性,对肺脏有一定的保护作用。
Objective To investigate the changes of serum levels of MMP-9 and the pathological changes of lung tissue in rats with sepsis induced by CLP ,and to explore the protection effect and action mechanism of Xuebijing injection . Methods Forty five male SD rats were randomly divided into 3 groups: blank group,NS group and Xuebijing group .Serum MMP-9 levels and the ratio of lung wet/dry weight were determined .The pathologic changes of lung were observed by means of light microscopy and transmission electron microscopy .Results As compared with that in blank group ,the ratio of lung wet/dry weight in NS group and Xuebijing group was significantly increased ( P &lt;0.01),however,as compared with that in NS group,the ratio of lung wet/dry weight in Xuebijing group was significantly decreased ( P &lt;0.01).The serum levels of MMP-9 in NS group and Xuebijing group were significantly higher than those in blank group ( P &lt;0.01).As compared with those in NS group,the serum levels of MMP-9 in Xuebijing group was significantly decreased ( P &lt;0.01).The lung tissue structures and vascular endothelial cell structures in blank group were basically normal ,however ,obvious inflammatory reaction and vascular endothelial cell structure injury were found in NS group , which in Xuebijing group were obviously relieved , however,which were severer than those in blank group .Conclusion Xuebijing injection can reduce MMP-9 levels, as a result,which can relieve lung vascular endothelial cell injury and improve pulmonary vasopermeability ,so Xuebijing injection has certain protection effect on lungs .
