摘要
为了研究鸡蛋清中卵转铁蛋白的分离提取工艺,采用硫酸铵沉淀和离子交换层析分离卵转铁蛋白,用聚丙烯酰胺凝胶电泳(SDS—PAGE)检测其纯度。鸡蛋清原液用Tris—HCl缓冲液稀释5倍,4℃下静置6h以上,采用50%~80%硫酸铵分级沉淀和Q—Sepharose FF离子交换层析分离卵转铁蛋白。结果显示,分离提取的卵转铁蛋白纯度达到96%,回收率为88.63%,且其具有生物活性,对大肠杆菌的抑菌率为48.84%。表明采用硫酸铵分级沉淀和Q—SepharoseFF离子交换层析法分离鸡蛋清中卵转铁蛋白可行。
To study separation processing of ovotransferrin from egg white proteins, ovotransferrin was separated by ammonium sulfates and anion-exchange chromatography. Purity of ovotransferrin was assayed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDSPAGE). Diluted egg white with five times volume of Tris HC1 buffer solution, deposited for at least 6 hours at 4 ℃ ,was treated by fractional precipitation with ammonium sulfate(from 50% to 80%) and Q-Sepharose FF anion-exchange chromatography. It showed that the purity of ovotransferrin was 96% by SDS-PAGE detection,the recovery was 88.63%,and bacteriostasis rate against E. coli was 48.84%. Ammonium sulfate precipitation and Q-Sepharose FF ion exchange chromatography was feasible to separate ovotransferrin from egg white.
