摘要
目的建立高效液相-示差折光检测器(HPLC-RID)法测定蒜酶提取物中聚乙二醇(PEG)含量的方法,并进行方法学考察。方法高效液相色谱法色谱柱为TSK gel G3000PWXL(7.8 mm×300 mm,7μm),流动相为纯水,柱温25℃,流速0.7 mg/mL,检测器为示差检测器,进样量15μL进行PEG含量检测。结果 PEG在浓度为0.05~15 mg/mL时,呈良好线性,相关系数r=1;精密度实验峰面积RSD分别为0.32%(日内精密度)和1.35%(日间精密度);重复性实验峰面积RSD为0.35%;稳定性实验峰面积RSD为1.27%;低、中、高浓度回收率分别为101.077%、100.045%、99.213%,RSD值分别为1.05%、1.08%、1.58%。6批蒜酶提取物中PEG平均含量为456.4mg/g。结论建立的HPLC-RID法各项检测指标均符合要求,可用于测定蒜酶提取物中PEG含量。
Objective To establish a method for the determination of polyethene glycol(PEG)content in Alliinase extract by High performance liquid chromatography-refractive index dector(HPLC-RID).Methods HPLC column was TSK gel G3000PWXL(7.8 mm×300 mm,7μm),mobile phase was pure water,column temperature was 25℃,flow rate was 0.7mg/ml,detector was differential detector,injection volume 15μL.Results The PEG showed good linearity at a concentration of 0.05-15 mg/mL,and the correlation coefficient was r=1.The precision experimental peak area RSD was 0.32%(intraday precision),1.35%(daytime precision);repeatability experiment.The peak area RSD was 0.35%;the stability experimental peak area RSD was 1.27%;the low,medium and high concentration recovery rates were 101.077%,100.045%and 99.213%,respectively,and the RSD values were 1.05%,1.08%and 1.58%,respectively.The average PEG content in the extract was 456.4 mg/g.Conclusion The established HPLC-RID method can meet the requirements and can be used to determine the PEG content in garlic extract.
