摘要
目的:建立HPLC法同时测定丹参药材中丹酚酸B、丹参酮ⅡA含量的方法。方法:采用Waters C18色谱柱(150mm×3.9mm,5μm),以0.3%甲酸水溶液-乙腈为流动相进行梯度洗脱,流速1.0mL/min,丹酚酸B、丹参酮ⅡA的检测波长分别为286nm、270nm。结果:丹酚酸B、丹参酮ⅡA分离良好,两成分质量浓度与峰面积在测定范围内线性关系良好,重现性好,平均加样回收率分别为100.54%和100.58%;8批不同产地丹参药材中丹酚酸B占药材量的平均含量为5.371 7%,丹参酮ⅡA占药材量的平均含量为0.403 2%。结论:本研究所建立的HPLC法稳定可靠,简便易行,可同时用于丹参中丹酚酸B、丹参酮ⅡA的测定,为丹参质量标准的提升提供了一定的理论依据。
Objective:To establish an HPLC method for simultaneous determination of Salvianolic Acid B and Tanshinone Ⅱ A in Salvia Miltiorrhiza Bunge. Methods:An HPLC in combination with PDA detector was applied with a WATERS C18 ( 150mm×3.9mm,5μm ) column by gradient elution using acetonitrile 0.3 % formic acid solution as the mobile phase. The flow rate was lmL/ rain and the detection wavelength was set at 280nm and 270nm. Results : Excellent chromatographic separation was achieved for two compounds with good linearity over the studied concentration ranges. And the recovery rates were 100.54% and 100.58%. Conclusion:The HPLC method developed for simultaneous determination of two compounds is simple and valid. It can be used for quality evaluation and quality control of Salvia Miltiorrhiza Bunge .
