溃疡性结肠炎外周血RORC和Foxp3 mRNA表达水平的变化及黄芩苷的干预作用 被引量:3

摘要 目的:本研究旨在探讨活动期UC患者外周血单个核细胞RORC和Foxp3 mRNA及相关细胞因子并予以不同浓度的黄芩苷干预的作用。方法:应用Q-PCR检测转录因子RORC和Foxp3 mRNA的表达,用ELISA法检测血清IFN-r、IL-6、IL-10、TGF-β水平及体外黄芩苷干预下的检测上述指标。结果:研究发现IFN-r、IL-4、IL-5、IL-10和TGF-β1与IBS-D组和健康对照组均有不同程度的升高,IL-6较IBS-D组和健康对照组稍降低。体外不同溶度的黄芩苷可引起IFN-r、IL-5、IL-6明显降低,IL-4和IL-10升高,以20μmol和40μmol浓度更明显。与健康对照组和IBS-D组相比较,UC患者的RORC和Foxp3mRNA明显增高,差异有显著性(P<0.05)。体外黄芩苷干预下,40μmol浓度干预下的RORC和Foxp3均明显下降,RORγt/FOXP3比值降低,间接调节了Th17/Treg分化的平衡。结论:较高浓度黄芩苷干预对Foxp3下降的作用更明显,使RORγt/FOXP3比值降低,间接调节了Th17/Treg分化的平衡,黄芩苷作为黄芩汤的重要组成成分之一,可能通过调节免疫平衡而缓解溃疡性结肠的炎症反应,为深入研究UC的发病机制及新药开发提供理论基础。 Objective:The purpose of this study is to explore the role of active UC patients'peripheral blood mononuclear cells RORC and Foxp3 mRNA and related cytokines and different concentrations of baicalin. Methods : Q - PCR detection of transcrip- tion factor RORC and Foxp3 mRNA expression were detected by ELISA in serum IFN - r, IL -6, IL - 10, TGF - 131 levels and the indicators were detected in vitro intervened by baicalin. Results:The study found that IFN -r, IL -4, IL- 5, IL- 10 and TGF - 131 and those of IBS - D group and the healthy control grouP were increased in varying degrees, IL - 6 compared with that in IBS - D and healthy control groups was slightly reduced. In vitro, different solubilities of baicalin caused IFN - r, IL - 5 and IL - 6 significantly reduced and IL -4 and IL - 10 increased, 20 μmol and 40μmol were more obvious. UC patients' RORC and Foxp3mRNA were significantly higher compared with healthy controls and IBS -D group, and the difference was significant(P 〈 0. 05 ). Baicalin intervention in vitro with concentration of 40μmol can significantly reduce RORC, Foxp3 and RORγt/FOXP3, in- directly regulate the balance of the Thl7/Treg differentiation. Conclusion:The high concentration of baicalin intervention has more obvious role on reducing Foxp3 and RORγt/FOXP3 ratio, and indirectly regulates the Thl7/Treg differentiation balance. Baicalin, as one of the important components of HQT, may regulate the immune balance and ease the ulcerative colon inflammation reac- tion, providing a theoretical basis for in - depth study of the pathogenesis of UC and the development of new drugs.
