摘要
建立了高效液相色谱法测定发酵液中色氨酸和酪氨酸含量的方法。采用Agilent C18柱(250mm×4.6mm,5μm ),以0.1mol/L醋酸钠(pH 4.0±0.05)-甲醇(90∶10)为流动相,流速为1.0mL·min-1,紫外检测器的检测波长为280nm,柱温30℃,结果显示,色氨酸的线性范围为50~500μg·mL-1,相关系数为r=0.9985,平均回收率为99.3%(n=4);酪氨酸的线性范围为20~2000μg·mL-1,相关系数为r=0.9991,平均回收率为100.2%(n=4)。该方法无需衍生,直接测定,简便,准确,用于发酵液中色氨酸和酪氨酸含量的测定,结果令人满意。
To establish HPLC method for the determination of fermentation liquid ammonia acid tryptophan and tyrosine. Agilent C18 column (250mm ×4.6mm,5μm )was used. The mobile phase was 0.1mol/L sodium acetate (pH 4.0 ±0.05)-Methanol(90∶10). The flow rate was 1.0mL·min-1 and detection wavelength was 280nm. Column temperature was 30℃. The results showed that tryptophan linear between 50~500μg·mL-1. The correlation coefficients was r=0.9985. The average recoveries was 99.3%(n=4). Tyrosine was linear between 20~2000μg·mL-1. The correlation coefficients was r=0.9991. The average recoveries was 100.2%(n=4). So drew the conclusions:Direct determination method without the need of derivative is simple,accurate, for the determination of content of fermentation liquid tryptophan and tyrosine.
