摘要
建立了HPLC法同时测定发酵液中洛伐他汀及其开环酸的含量。采用C8柱,流动相为10mmol/L磷酸-乙腈(40∶60),检测波长238nm。洛伐他汀及其开环酸分别在0.012~0.096mg/ml和0.01~0.08mg/ml范围内线性关系良好,平均回收率分别为99.8%和98.7%。
A HPLC method was established for the determination of lovastatin and lovastatin acid in the fermentation broth. A Ca column was used with the mobile phase of 10mmol/L phosphoric acid-acetonitrile (40 : 60), at the detection wavelength of 238nm. The calibration curves of lovastatin and lovastatin acid were linear in the ranges of 0.012 - 0.096mg/ml and 0.01 - 0.08mg/ml, respectively. The average recoveries were 99.8% and 98.7%, respectively.
