The impact of LPS on the expression of aquaporin-1 and its functions in human intestinal microvascular endothelial cells

摘要 目的:通过观察脂多糖(LPS)对人肠黏膜微血管内皮细胞水通道蛋白-1(AQP1)表达及mRNA水平的影响,探讨水通道蛋白在脓毒症病人肠微血管内皮细胞通透性改变中的意义。方法:在体外培养的人肠黏膜微血管内皮细胞(HIMEC),根据LPS的浓度分为四组:即低浓度组(LPS 0.1 mg/L)、中浓度组(LPS 1 mg/L)、高浓度组(LPS 10 mg/L)和对照组(只加培养液),培养6、12和24 h后,应用细胞免疫荧光RT-PCR法测定AQP1表达及其mRNA的水平。结果:与对照组相比,LPS作用于肠微血管内皮细胞后,AQP1蛋白及其mRNA的表达均下降(P<0.05),并且与LPS的浓度和作用时间呈相关性趋势。结论:LPS刺激HIMEC后,AQP1出现蛋白表达量下降及其mRNA水平降低。AQP1可能与急性肠损伤后肠黏膜微血管内皮细胞的通透性增加有关。 Objective: To explore the significance of AQP in permeability changes of intestinal microvascular endothelial cell for patients with sepsis, by observing the impact of LPS on the expression of aquaporin-1 and m-RNA level in human intestinal microvascular endothelial cells. Methods: The cultured human intestinal microvascular endothelial cells (HIMECs) were divide into four groups accord- ing to the concentration of LPS. After culture for 6 h, 12 h, 24 h, the expression of AQP1 and mRNA level were measured by immunofluorescence and RT-PCR. Results: Compared with the control group, AQP1 protein and its mRNA expression decreased (P 〈 0.05), and had a certain concentration and time-dependent trend. Conclusion: After the stimulation of LPS on HIMECs, AQP1 protein and its mRNA expression may decrease, and it may be related to increased intestinal microvascular endothelial cells permeability after acute intestinal injury.
