摘要
利用碱-醇法从酒糟中提取蛋白质。根据单因素试验结果选取了碱-醇比、液料比、反应温度、反应时间作为影响蛋白提取率的因素。采用四因素三水平响应面分析法对酒糟蛋白提取工艺进行优化研究,确定了碱-醇法提取的最佳工艺参数为碱-醇体积比2.22∶1,液料比31.91∶1,反应温度33.86℃,反应时间75.4min。在此条件下酒糟蛋白的提取率达到10.182 7%。由酒糟蛋白浓缩液配制的蛋白泡沫灭火剂其发泡倍数、析液时间、灭火时间、抗烧时间等指标均达到GB15308-2006《泡沫灭火剂》标准要求。
Alkali-alcohol extraction method was used to extract protein from distiller's grains. The four variables that influence extraction efficiency were the ratio of alkali and alcohol, the ra- tio of extraction solvent and distiller's grains, temperature and time determined by the one-factor-at-a-time method. Response surface analysis was used to optimize the extraction conditions among the four variables at three levels on extraction efficiency. The optimum ratio of alkali and alcohol was 2.22 : 1, the ratio of extraction solvent and distiller's grains was 31.91 : 1, tem- perature was 33. 86℃ and time was 75.4 min. Under these conditions, the extraction efficiency of protein was 10. 182 7%. The properties of protein foam extinguishing agent made from hydrolyzed protein reached the national standard of the People's Republic of China (GB 15308- 2006, Foam extinguishing a- gent).
