摘要
从水稻田和玉米田土样中分别分离适用于水田作物和旱田作物的硅酸盐细菌,对能够在亚历山大罗夫培养基上生长且符合硅酸盐细菌菌落特征的细菌菌株进行菌种鉴定,然后测定在不同供氧条件下菌株的解硅、解钾能力。结果表明,从水稻田土样中分离得到菌株2株,从玉米田土样中分离得到菌株1株,经鉴定均属于硅酸盐细菌的一种———胶冻样类芽孢杆菌(Paenibacillus mucilaginosus)。在不同供氧条件下,这3株菌株均具有一定的解硅、解钾活性,供氧充足的条件下菌株h-5的解硅活性最高,可以使发酵液中水溶性硅含量达到4.5μg/mL以上,菌株s-4解钾活性最高,可以使水溶性钾含量达到46.73 mg/L以上。分离自水田的2株菌株在微氧条件下仍能保持较高的解硅、解钾活性,具有作为生物肥料的出发菌株应用于水田作物的较大潜力。
From the soil samples of paddy field in Hunan area and corn field in Hebei area, several silicate bacteria strains were isolated Then the strains were identified by morphological and culture features observation. The activity of releasing sili- con and potassium of these strains on different levels of oxygen dissolution were determined. The results showed that two strains and one strain were obtained from paddy soil and corn field soil, respectively. These three strains were all identified as Paenibacillus mucilaginosus, a kind of silicate bacteria. These three strains had a certain level of activity of releasing sili- con and potassium. Among them, strain h-5 had the highest activity of releasing silicon, the content of water soluble silicon in its fermentation broth could reach over 4.5 μg / mL. Strain s-4 had the highest activity of releasing potassium, the content of water soluble potassium in its broth were over 46.73 mg / L. The activity of releasing silicon and potassium of two strains obtained from paddy soil were stabler on different levels of oxygen dissolution than that of other strain. On the condition of low oxygen dissolution, the two strains obtained from paddy soil could still keep a relatively high activity of releasing silicon and potassium.
