摘要
通过对固氮、释磷能力以及对层出镰孢菌抗病能力的测定,筛选得到固氮菌N3(Arthrobacter sp.ZSH N3)、水解土壤有机磷细菌Y1(Enterobacter asburiae ZSH Y1)、释放不溶性无机磷细菌W3(Enterobacter hormaechei ZSH W3)和生防菌KY2(Bacillus methylotrophicus KY2)。采用单因素试验、响应面试验,以活菌数为指标,优化玉米秸秆固态发酵工艺的参数,在温度30.4℃、初始pH 7.8、初始含水量100%、接种量15.5%、接种间隔时间24 h、发酵时间120 h,菌肥中N3、Y1、W3和KY2实际有效活菌数分别达到2.27×10^9、3.11×10^9、7.10×10^8和3.21×10^9 CFU·g^-1,总氮和有效磷含量分别为原材料的6.07倍和11.43倍。盆栽试验表明,菌肥能够显著促进烟草的光合作用,并有效防治层出镰孢菌引起的枯萎病。
The nitrogen-fixing bacteria N3(Arthrobacter sp.ZSH N3),organic phosphorus solubilization bacteria Y1(Enterobacter asburiae ZSH Y1),inorganic phosphorus solubilization bacteria W3(Enterobacter hormaechei ZSH W3)and bio-control bacteria KY2(Bacillus methylotrophicus KY2)were screened by measuring the ability of nitrogen fixation,phosphorus solubilization and resistance to Fusarium proliferatum.The parameters of solid state fermentation technology of corn stalks were optimized by single factor and response surface experiments with the total number of live bacteria as index.The optimal conditions predicted by response surface experimental results were fermentation temperature of 30.4℃,initial pH of 7.8,initial water content of 100%,inoculum of 15.5%,inoculation interval time of 24 h and fermentation time of 120 h.Under the optimal conditions,the total actual effective viable bacteria number of N3,Y1,W3 and KY2 in the microbial fertilizer were up to 2.27×10^9 CFU·g^-1,3.11×10^9 CFU·g^-1,7.10×10^8 CFU·g^-1 and 3.21×10^9 CFU·g^-1,respectively.The content of total nitrogen and available phosphorus were 6.07 and 11.43 folds compare with the raw materials.The pot experiments showed that the microbial fertilizer could significantly promote the tobacco photosynthesis and effectively inhibited the wilt caused by Fusarium proliferatum.
