摘要
目的观察脑瘤消胶囊对环己亚硝脲(CCNU)所致S180肉瘤小鼠骨髓造血功能与肝肾功能损害的预防治疗作用。方法在建立S180肉瘤小鼠模型的基础上,设荷瘤空白对照组(以生理盐水灌胃)、环己亚硝脲组(以40mg/kg的CCNU灌胃)及CCNU+小剂量NLX组、CCNU+中剂量NLX组、CCNU+大剂量NLX组(分别以2.5%、5%、10%的脑瘤消胶囊混悬液和40mg/kg的CCNU灌胃)和平消胶囊+CCNU组(以5%平消胶囊混悬液和40mg/kg的CCNU灌胃);检测外周血象与肝肾功能、骨髓粒细胞、红细胞数,测定脾脏指数和胸腺指数。结果脑瘤消胶囊在500、1000、2000mg/kg剂量并联合CCNU时,与CCNU单用组比较,可对抗环己亚硝脲引起的荷瘤S180小鼠外周血白细胞和网织红细胞减少(P<0.01),可使荷瘤S180小鼠骨髓粒细胞比值上升和骨髓红细胞比值回落(P<0.01),可减轻CCNU引起的小鼠肝功能损害(P<0.05),对肾功能无明显影响(P>0.05);可升高荷瘤S180小鼠胸腺和脾脏指数(P<0.01)。结论脑瘤消胶囊可对抗细胞毒药物环己亚硝脲所引起的骨髓造血功能和肝功能损害及免疫抑制作用。
Objective To observe the prevention treatment of the NLX Capsule on injury of bone marrow, liver and kidney function induced by cyclohexyl nitrosourea （CCNU）in S180-bearing mice. Methods The S180-bearing mouse were randomLy divided into 6 eaqual goups： control group（gavaged by saline）, cyclohexyl nitrosourea group （gavage by 40mg/kg of CCNU）, CCNU ＋ low-dose NLX group, CCNU ＋ middle-dose NLX group, CCNU ＋ high-dose NLX group （gavage by 2.5%, 5%, 10% of the NLX capsule suspension and cancellation of CCNU 40mg/kg）, and Pingxiao capsule ＋ CCNU group（gavaged by 5% Pingxiao capsule suspensions and 40mg/kg of CCNU）. Peripheral blood, liver and kidney function, bone marrow granulocyte and erythrocyte count were detected, spleen and thymus index were also measured. Results In S180-bearing mice, NLX capsule with 500, 1000, 2000 mg/kg plus CCNU, could antagonize white blood cells and reticulocytes reduce induced by CCNU（P〈0.01）, promote bone marrow granulocyte ratio increased and red blood cell ratio declined（P〈0.01）, reduce liver damage（P〈0.05）, renal function had no significant effect（P〉0.05）, can be elevated tumor thymus and spleen index in SI80- bearing mice（P〈0.01）. Conclusion NLX capsules against cytotoxic cyclohexyl nitrosourea induced bone marrow function and liver dysfunction and immune inhibition.
