聚酰胺树脂纯化昆仑雪菊总黄酮的工艺研究 被引量:5

摘要 目的优选聚酰胺树脂分离纯化昆仑雪菊总黄酮的工艺条件。方法采用紫外分光光度法,以总黄酮含量为考察指标,研究在静态和动态条件下,聚酰胺树脂吸附和洗脱总黄酮的行为,筛选出聚酰胺树脂精制雪菊总黄酮的最优工艺参数。结果雪菊总黄酮最佳纯化工艺条件为:60~100目聚酰胺对雪菊总黄酮吸附与解吸较好;清洗液最佳pH值为5;乙醇用量为11倍柱体积;乙醇浓度为70%,在该纯化条件下,总黄酮纯度由粗提物22.56%提高到68.04%。结论聚酰胺树脂对雪菊总黄酮的纯化效果好,成本低,可工业化推广。 Objective To optimize purification technique for total flavonoids from Kunlun Chrysanthemum by polyamide resin. Methods With the content of total flavonoids as index, by using UV spectrophotome- try,adsorption and elution pattern of polyamide resin for total flavonoids was investigated under static and dynamic conditions, and selected optimum technique parameters of purification technology for total fla- vonoids by polyamide resin. Results The best parameters for separating the total flavonoids of Kunlun Chrysanthemum: Adsorption and desorption abilities of 60~100 mech polyamide for total flavonoids were the best of the three polyamides; Optimum pH value of the cleaning liquid 5; the amount of ethanol is 11 column volumes; ethanol concentration of 70%. Under the purification condition, the content of total fla- vonoids could reach 68.04% from 22.56%. Conclusion Because of the good effect and low cost, Polyamide resin for purification of total flavonoids Kunlun Chrysanthemum can be widely used in practice.
