摘要
试验选择体重3.23±0.63 kg奇卡兔20只,单独或联合应用组胺和4-氨基吡啶(4-AP)建立兔瘙痒模型。将兔随机分为4组,皮下注射造模药物后记录搔抓反应次数和搔抓潜伏期,检测注射药物前后小鼠血清中的组胺、IFN-γ和IL-6的浓度。结果显示:组胺和4-AP联合运用后搔抓潜伏期显著缩短,40 min内搔抓次数均极显著增加(P<0.01)。组胺致瘙痒模型组的兔血清组胺浓度升高,与其他3组相比差异极显著(P<O.01);其他2个细胞因子的浓度在各组之间差异均未达到统计学显著水平。联合应用组胺和4-AP建立的瘙痒模型效果优于组胺和4-AP单独作用。
Twenty rabbits, body weight 3.23 ±0.63 kg, were selected for the study. Histamine and 4 - aminopyridine (4 - AP) was solely or combinely administrated to establish rabbit itching model. Rabbits were randomly divided to 4 groups. Scratching incubation period (ICP), scratching numbers within 40 minutes were recorded after hypodermic injection with drug administration. Concentration of histamine, interferon - γ' ( IFN - γ) and interleukin -6 (IL- 6) in plasma were detected with the ELISA kit before and after drug administration. The result showed that ICP was significantly decreased and scratching numbers were significantly increased after combination use of histamine and 4 - AP compared with non - drug administration ( P 〈0. 01 ). The serum concentrations of histamine decreased in the histamine group. It was significantly higher than the other three groups(P 〈0.01 ). The serum concentrations of IFN -γand IL - 6 had no significant difference among these four groups ( P 〉 0.05 ). Pruritus model could be established by combination ues of histamine and 4 - AP.
