摘要 为了实现红叶乌桕优良种源及变异个体的种质保存和快速繁殖,以带腋芽茎段、茎尖、叶片为外植体,选择不同接种时期、不同基本培养基、不同激素种类和浓度,进行了红叶乌桕的组织培养技术研究。结果表明:6月中旬取半木质化茎段,用0.1%HgCl2灭菌5 min效果较好,外植体接种成功率达79.5%;MS+KT 0.5 mg/L+IBA 0.5 mg/L培养基适于试管苗继代增殖,20~25天继代一次长势良好;试管苗在MS+IBA 0.5 mg/L+KT 0.5 mg/L和1/2MS+IBA 0.5 mg/L培养基中生根效果较好,生根率为97.62%~100%,平均主根数为9.21~13.03条/株。 For germplasm conservation and rapid propagation of Euphorbia cotinifolia, the stem-segment with axillary buds, stem tips and leaflet was taken as explants to be studied on different inoculating, medium, hormone and its concentration for tissue culture system. The results showed that, the optimizing sterilization method for stem-segment with axillary buds in the second ten days of June and explants treated with 0.1%HgCl2 for 5 min was the better. The percentage of sterile explants could arrive at 79.5%. It was suggested that MS+KT 0.5 mg/L+IBA 0.5 mg/L to be desirable treatment for subculture and multiplication, with 2.45 times multiplication rate after a 20-25 days cultivation. The media with MS+IBA 0.5 mg/L+KT 0.5 mg/L and 1/2MS+IBA 0.5 mg/L could be used for the rooting induction. The rooting rate was 97.62%-100%. The average number of rootlet for the test-tube plantlets was 9.21-13.03.
