摘要
目的研究复方中药天芪航力片对高+Gz应激大鼠心肌线粒体功能的影响。方法 72只雄性SD大鼠随机分为空白对照组、应激对照组、高+Gz应激组、复方低剂量组、复方中剂量组和复方高剂量组,每组12只。各组大鼠均灌胃给药,给药体积为20ml/kg,1次/d,连续14d。其中,复方低、中、高剂量组分别按0.75、1.5和3.0g/kg给予天芪航力片制剂,空白对照组、应激对照组和高+Gz应激组给予等量生理盐水。第15天后4组大鼠进行离心机暴露(+10Gz,5min)。观察各组大鼠心肌线粒体呼吸功能、抗氧化能力和ATP酶活性的变化。结果与空白对照组和应激对照组相比,高+Gz应激组大鼠心肌线粒体Ⅲ态呼吸(ST3)和呼吸控制率(RCR)明显降低(P<0.01),Ⅳ态呼吸(ST4)明显升高(P<0.05),线粒体超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)及Na+-K+-ATP酶活性也明显降低(P<0.05或P<0.01),而丙二醛(MDA)含量显著增加(P<0.01)。复方中药预处理可提高+10Gz暴露后大鼠线粒体ST3和RCR,增强线粒体抗氧化酶和ATP酶的活性,复方高剂量组大鼠心肌线粒体ST3、RCR和抗氧化酶、ATP酶活性显著高于高+Gz应激组(P<0.05或P<0.01),SOD活性甚至高于空白对照组(P<0.05),而MDA含量则明显降低(P<0.01)。结论复方中药天芪航力片可以改善高+Gz应激大鼠心肌线粒体呼吸功能,提高线粒体抗氧化能力和ATP酶的活性,对心肌线粒体损伤具有保护作用。
Objective To evaluate the effect of compound Tianqi Hangli tablet on the function of myocardial mitochondria in rats after high +Gz stress exposure. Methods Seventy-two male SD rats were randomly divided into 6 groups (12 each): blank control group, stress control group, high +Gz stress group, low dose compound group, medium dose compound group, and high dose compound group. Tianqi Hangli tablet was administered orally to rats once a day for 14 days. Low-, medium- and high dose compound group were fed with compound Tianqi Hangli tablet at doses of 0.75, 1.5 and 3.0g/kg, respectively. Rats in the other three groups were fed with equal volume of saline. Experimental rats were exposed to high +Gz at the 15th day. The respiratory function, antioxidant capacity and ATPase activity of myocardial mitochondria were detected after +Gz exposure. Results Compared with blank control group and stress control groups, the state 3 respiration (ST3) and respiratory control ratio (RCR) of myocardial mitochondria of rats in high +Gz stress group significantly decreased (P〈0.01), the state 4 respiration (ST4) significantly increased (P〈0.05), the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and Na+-K+-ATPase decreased (P〈0.05 or P〈0.01), but the content of malondialdehyde (MDA) increased (P〈0.01). However, compound preconditioning increased ST3 and RCR, and enhanced the activities of antioxidant enzymes and ATPase in rats suffering +10Gz exposure. The ST3, RCR and the activities of antioxidant enzymes and ATPase of rats in high dose compound group were significantly higher than those in high +Gz stress group (P〈0.05, P〈0.01), SOD activity was even higher than those in blank control group (P〈0.05), and the MDA content declined significantly (P〈0.01). Conclusion Compound Tianqi Hangli tablet may have a protective effect on myocardial mitochondria in rats after high +Gz stress exposure via ameliorating respirator
