摘要
采用响应面法优化生物发酵法制备谷氨酸的培养条件。以谷氨酸产酸率及糖化转化率为考察指标,通过Plackett-Burman(PB)及中心组合设计(CCD)筛选并优化影响谷氨酸发酵的显著因素,建立回归模型,通过响应面数据分析,确定生物发酵法制备谷氨酸的培养条件为:口服葡萄糖181.69 g/L、玉米浆4.0 g/L、糖蜜2 g/L、K2HPO41.5 g/L、KCl 0.04 g/L、MgSO4·7H2O 0.55 g/L,接种量6%,发酵液的装液量为29.7 mL/1000 mL,在此条件下,其谷氨酸产率和糖酸转化率分别达到了12.32%和64.3%,较优化前分别提高了42.5%和28.6%。
Response surface method was used to optimize fermentation conditions for the production of glutamic acid. In order to obtain high glutamic acid yield and transformation efficiency, the effect of fermentation conditions on glutamic acid production was studied using Plackett-Burman and central composite design analysis software. A mathematical regression model was established using response surface methodology. The results indicated that the composition of the optimized medium was: oral glucose 181.69 g/L, corn steep liquor 4 g/L, molasses 2 g/L, K2HPO41.5 g/L, KCI 0.04 g/L, MgSO4.7H20 0.55 g/L, inoculums size 6% with a medium loading of 29.7 mL/1000 mL. Under this conditions, the glutamic acid concentration and yield reached 12.32% (w/v) and 64.3%, which were increased by 42.5% and 28.6%, respectively.
