摘要 目的 :观察体外培养的人牙乳头间充质细胞 (dentalpapillamesenchymalcell,DPMC)矿化特征及甲状旁腺激素 (parathyroidhormone ,PTH)对人DPMC矿化特征的影响 ,以深入研究成牙本质细胞的分化和牙本质形成。方法 :第 4代人DPMC在含 33 .3nmol/LPTH的矿化液中连续培养 35d后 ,进行组织学染色、透射电镜(TEM)和扫描电镜 (SEM)观察。用不含PTH的矿化液进行对照。结果 :人DPMC可出现复层生长和形成矿化结节 ;vonKossa染色提示细胞结节中有明显的钙盐沉积 ;细胞可出现与成牙本质细胞相似的超微结构特征 ,并出现典型的矿化影像。加入PTH显著增加细胞的矿化结节 ,并出现大量基质小泡。结论 :体外培养的人DPMC的矿化过程可能与基质小泡有关。PTH能显著提高人DPMC的矿化程度 。 AIM: Observation of the effects of parathyroid hormone on the calcification ability of cultured human dental papilla mesenchymal cell (DPMC). METHODS: Two groups of the fourth passage human DPMC were cultured for 35 days and observed by transmission electron microscope(TEM), scanning electron microscope(SEM) and light microscope(LM). The control group was cultured in DMEM medium supplemented with 15% FCS, 50mg/L ascorbic acid and 2mmol/L β-glycerophosphate. The PTH group was cultured in the medium of control group containing 33.3nmol/L PTH. RESULTS: The cell presented mineralized nodules as well as matrix vesicles and had the same ultrastructure with odontoblasts under TEM. The addition of PTH in the medium caused not only a significant increase in the number of nodules but also the formation of many matrix vesicles. CONCLUSION: The results suggest that cultured human DMPC have the calcification ability which may be related to the matrix vesicles and PTH enhances the calcification ability of cultured human DPMC. [
