摘要 目的 以肿瘤抗原P815AB抗原肽为模式抗原 ,研究CpG寡聚脱氧核苷酸 (CpGODN)对该肿瘤抗原表位肽的免疫佐剂效应。方法 用 [3H ] TdR掺入法研究CpGODN体外刺激小鼠脾细胞增殖效应 ,用ELISPOT和 [3H] TdR释放法检测不同构成形式的疫苗在体诱发特异性CTL应答的效应。结果 ODN182 6在体外可刺激小鼠脾细胞显著增殖。经 4次免疫 ,在“肽 +ODN182 6 +IFA”组中 ,4/6的小鼠体内出现了特异性CTL应答 ;在“肽 +ODN182 6 +PBS”组 ,有 3只小鼠在完成 4次免疫前先后死亡 ,余 3只中有 1只体内也可检测到相应CTL反应 ;而对照“肽 +IFA”组 ,6只小鼠体内均未诱发出特异性CTL应答。结论 CpGODN对P815AB表位肽有较为理想的免疫佐剂效应 ,但疫苗构成形式对其免疫效果有着决定性的作用 ,“肽 +ODN182 6 +IFA”是本研究中最有效的疫苗形式 。 Objective To study the adjuvant effects of CpG containing oligodeoxynucleotide (CpG ODN) on the immunization of antigen peptide P815AB. Methods The proliferation of mice splenocytes induced by CpG ODN in vitro was assessed by [ 3H]thymidine incorporation; the specific CTL responses of mice immunized with different forms of peptide vaccines were measured with ELISPOT and [ 3H]thymidine release assay. Results ODN1826 could induce the significant proliferation of mice splenocytes in vitro . After four immunizations, measurable CTL responses against P815AB could be induced in four of six mice immunized with “P815AB+ODN1826+IFA”, and in one of three mice remaining alive immunized with “P815AB+ODN1826+PBS”, but none of six mice immunized with “P815AB+IFA”. Conclusion CpG ODN is an efficient adjuvant of specific CTL responses for mice immunized with tumor specific peptide P815AB, and “P815AB+ODN1826+IFA” is the most potent form of vaccine in this study. However, CpG ODN may cause harmful side effects to some extent.
