目的检测子宫内膜异位症(EMS)患者人白细胞抗原-G(HLA-G)基因第8外显子3'-非翻译区14 bp插入/缺失多态性和子宫内膜组织(在位和异位内膜)HLA-G蛋白表达水平,分析它们与EMS的关系。方法利用多聚酶链反应-序列特异性引物法(PCR-SSP)检测92例EMS患者(EMS组)和90例非EMS患者(对照组)的HLA-G基因第8外显子3'-非翻译区14 bp插入/缺失多态性的基因型分布,免疫组织化学染色检测其中30例EMS患者在位及异位内膜和30例非EMS患者在位内膜HLA-G蛋白表达水平。结果 HLA-G 14 bp插入/缺失多态性基因频率在EMS组和对照组比较差异均无统计学意义(P均>0.05);HLA-G蛋白在EMS组异位内膜表达明显高于对照组(P<0.05),但在EMS组在位内膜和异位内膜之间的表达比较差异无统计学意义(P>0.05);进一步分析HLA-G阳性表达程度与EMS患者的HLA-G 14 bp插入/缺失多态性基因型分布之间的关系,组间比较未见明显差异(P均>0.05)。结论 EMS发病可能与内膜组织中HLA-G的异常表达有关,而与HLA-G基因第8外显子3'-非翻译区14 bp插入/缺失多态性无关。
Objective To determine the association of HLA-G 14 bp insertion/deletion polymor- phism and endometrium HLA-G expression, and analyze their correlation with endometriosis (EMS). Methods The polymeric chain reaction/sequence specific primer (PCR-SSP) technique was performed to detect the HLA-G 14bp deletion/insertion polymorphism in peripheral blood samples from patients with (n = 92 ) and without Endometriosis (n = 90 ). HLA-G expression was detected among eutopic endometrial biopsies from women without ( n = 30) and with ( n = 30) endometriosis, as well as ovarian endometriotic lesions ( n = 30 ). Results No significant difference was observed in the distribution of -14 bp insertion/deletion genotype nor the number of -14 bp insertion alleles between controls and the EMS group. There was a statistically significant HLA-G expressionwas detected in glandular cells, with higher expression in eetopic endometrium with EMS than that without EMS, with statistical significan. However, no significant difference was noted in HLA-G expression between the glandular cells of eutopic and ectopic endometrium from patients with EMS. Also, no statistical difference was observed in the levels of the expression of HLA-G protein among-14 bp insertion/deletion genotypes. Conclusions The abnormal expression of HLA-G indicates that HLA-G may play an important role in the pathogenesis of EMS, but be independence of HLA-G 14 bp ins/del polymorphism.
