摘要
目的:研究茶枝柑皮多糖对H2O2诱导的PC12细胞氧化损伤的保护作用。方法:MTT法检测细胞存活率,建立H2O2诱导PC12细胞氧化损伤模型。比色法测定细胞内丙二醛(MDA)含量、超氧化物歧化酶(SOD)以及谷胱甘肽过氧化物酶(GSH-Px)的活性。结果:100μmol/L H2O2诱导PC12细胞4h,细胞呈现明显损伤形态,细胞内MDA含量升高,SOD和GSH-Px活性降低。茶枝柑皮多糖可明显改善PC12细胞损伤,显著降低细胞MDA含量,极显著提高SOD和GSH-Px活性。结论:茶枝柑皮多糖对H2O2诱导PC12细胞损伤具有明显的保护作用,其作用机制可能与提高PC12细胞的抗氧化酶活性有关。
Objective:To study the protective effect of polysaccharide from Citrus reticulata 'Chachi' Peel on H2O2-induced oxidative injury in PC12 cells. Methods.Cell viability was assayed by MTT method,the toxicity model was established by treating PC12 cells with H2O2. The level of MDA,SOD and GSH-Px were measured by spectroscopy, respectively. Results.After being exposed to 100μmol/L H2O2 for 4 hours,typical morphological characteristics of injury were observed, The level of MDA increased significantly,while that of SOD and GSH- Px decreased significantly in H2O2-induced group compared with that of the sham group. Polysaccharide from Citrus reticulata 'Chachi' Peel could decrease MDA content and increase SOD and GSH-Px activity significantly. Conclusions.Polysaccharide from Citrus reticulata 'Chachi' Peel had protective effect on PC12 ceils injury induced by H2O2 in culture. The protective effect might be related to the increased activity of antioxidant enzymes.
