摘要
目的研究结直肠癌(colorectal carcinoma,CRC)患者外周血中Th17细胞的含量及其效应分子白介素17(interleukin-17,IL-17)的表达情况,探讨Th17与CRC发生和发展之间的关系。方法采用流式细胞术和ELISA方法,检测25例Ⅰ-Ⅱ期CRC患者、18例CRCⅢ-Ⅳ期患者和10例健康志愿者(对照组)外周血中CD4+IL-17+细胞所占比例及血清中IL-17表达量的变化,分析Th17细胞与CRC患者各临床病理特征之间的关系。结果Ⅰ-Ⅱ期患者血清中IL-17表达量为(47.72±12.89)ng/L,Ⅲ-Ⅳ期患者IL-17表达量为(58.69±12.81)ng/L,均明显高于健康对照组(16.63±6.41)ng/L。Ⅰ-Ⅱ期患者外周血中CD4+IL-17+细胞占(2.24±0.54)%,Ⅲ-Ⅳ期患者外周血中CD4+IL-17+细胞占(2.65±0.63)%,均高于健康对照组(1.34±0.61)%。IL-17表达水平的上升与患者的性别和年龄无关(P>0.05),而与肿瘤的大小、淋巴结转移、Dukes分期及病理组织学分型明显相关(P<0.01)。结论 CRC患者外周血中Th17细胞含量和血清中IL-17的表达水平与肿瘤的进展相关,预示Th17细胞直接或间接参与了CRC发生和发展的调控。
Objective To study the frequency of peripheral blood Th17 cells and the expression levels of serum II.- 17 in patients with colorectal cancer (CRC),and to investigate the correlation between Th17 cells and the development of human CRC. Methods The concentration of IL-17 and the expression of Thl7 ceils in peripheral blood mononuclear cells (PBMC) were detected by ELISA and flow cytometry (FCM) respectively in 25 CRC patients in stage I-II , 18 CRC patients in stage III-IV and 10 healthy controls (HC group). Then analysed the correlationship between Th17 cells and clinical pathological characteristics of CRC. Results The concentration of serum IL-17 was (47.72±12.89) ng/L in patients with CRC stage I-II ,and (58. 69 ± 12.81) ng/L in patients with CRC stage III-IV, both of them were showed a higher level than that in HC group (16.63!6.41) ng/L. The frequency of Thl7 cells was (2.24±0.54)% in patients with CRC stage I -II ,2.65 ±0.63 % in patients with CRC stage III-IV, which possessed obviously difference with HC group (1.34 ± 0.61 % ) as well. The increased expression level of IL-17 associated with tumor size, lymphatic metastasis,Dukes staging and histopathological type (P〈0.01) and had no relations with sex or age of patients (P〉 0.05). Conclusion The expression level of serum IL 17 and the frequency of peripheral blood Thl7 cells associated with the development of CRC,which indicated Th17 cells may participate in regulating the development of CRC direct
