摘要
目的:观察苏木、苏木+黄芪对Lewis荷瘤小鼠肺癌转移过程脾Th17/Treg细胞及其平衡机制影响,为中药逆转肿瘤炎性微环境中免疫逃逸和抑制肿瘤转移机制提供实验依据。方法:复制C57BL/6小鼠Lewis肺癌动物模型,观察肺转移抑制率;流式细胞术检测各组小鼠脾Th17/Treg细胞百分比动态变化;RT-PCR检测各组小鼠CD4+T淋巴细胞Foxp3、RORγtmRNA动态表达水平。结果:从各组药物抑制肺转移率看出,益气活血药苏木+黄芪优于单纯活血药苏木(P<0.05),与环磷酰胺组相似(P>0.05);除苏木黄芪组外,各荷瘤组小鼠脾Th17与Treg细胞随时间迁移呈现出上升趋势,且苏木黄芪组较其余各组有显著差异(P<0.05),Th17/Treg比值亦随时间迁移呈现出上升趋势,与正常组比较均存在比值的平衡失调;Th17与Treg细胞关键转录分子RORγt、Foxp3呈现出与Th17和Treg细胞相应的动态改变。结论:益气活血药苏木+黄芪组在抑制肿瘤形成过程中的炎性微环境优于单纯活血药苏木组和环磷酰胺组;各组药物对肿瘤转移机制均有一定的抑制作用,但益气活血药优于单纯活血药和化疗药物。
Objective:To the effect of Huoxue drugs, YIqihuoxue drugs on the balancing of Th17/Treg cells in the process of metastasis of lung cancer and provide experimental basis for traditional Chinese medicine to reversing immune escap of tumor. Methods:The inhibition rate of lung metastasis in each group was observed; the dynamic percentage and ratio change of spleen Th17 and Treg cell were detected by flow -cytometry; the dynamic expression of Foxp3, RORγt mRNA of CD4+ T lymphocytes were detected by RT-PCR. Results:From the drugs inhibiting lung metastasis rate in each group, Yiqihuoxue drugs--Astragalus + lignum sappan were better than the Huoxue drug--lignum sappan ( P 〈 0. 05) , and similar with cyclophosphamide group(P 〉 0. 05) ;In addition to Astragalus + lignum sappan groups, spleen Th17 and Treg showed a rising trend, and Astragalus + lignum sappan groups had significant difference than other groups ( P 〈 0.05 ) ;The cell effector ( IL-17, IL-23, IFN) and cell key transcription molecule ( RORγt, Foxp3 ) present -ed the dynamic change corresponding to Th17 and Treg. Conclusion : Astragalus + lignum sappan groups in the inhibition of tumor immune tolerance and the formation of immune inflammatory response were better than the lignum sappan groups and cyclophosphamide groups; each group of drugs have certain effect on the mechanism of tumor metastasis, but Yiqihuoxue drugs were better than the Huoxue drug and chemotherapeutic drug.
