摘要
多酚氧化酶是植物体内广泛存在的铜结合酶,催化一元酚或多元酚氧化生成“黑色素”,是引起果蔬等农产品褐变的主要原因。通过对已克隆的植物多酚氧化酶基因的分析,设计了一对简并引物,PCR扩增了甘薯多酚氧化酶保守区的cDNA,序列分析结果表明,该cDNA含有591个核苷酸,编码197个氨基酸,与已发表的其它植物的多酚氧化酶保守区的cDNA具有很高的同源性。
Polyphenol oxidase(PPO) is a ubiquitous copper enzyme which catalyzes the oxi- dation of mono-and O-diphenols into O-diquinoies. PPO is believed to be primarily re- sponsible for deleterious browning of many fruits and vegetables. Conserved region of sweet potato PPO gene was cloned and sequenced through PCR using a pair of degenerte primer designed on the basis of analysis of cloned PPO gene sequences. Results show it consists of 591 nucleotides encoding 197 amino acids in all and has a very high homology to previously cloned PPO in other plants.
