摘要
目的:研究白细胞介素-10(IL-10)修饰的未成熟树突状细胞(immature dendritic cells,imDC)对大鼠同种异体角膜移植术后免疫耐受的影响。方法:用培养的供体来源的imDC及IL-10-imDC预处理受体,建立Wistar-SD大鼠穿透性角膜移植模型。供体Wistar大鼠18只;受体SD大鼠36只,受体鼠随机分为3组,每组均为12只。A组(对照组):受体鼠不经任何预处理即行角膜移植术;B组:角膜移植前3d,每只受体鼠经尾静脉注射供体的imDC,细胞数为2×106个;C组:角膜移植前3d,每只受体鼠经尾静脉注射用IL-10修饰的供体imDC,细胞数为2×106个。观察术后各组受体角膜植片的存活时间;术后第14d各组随机抽取4只鼠取术眼角膜行组织病理学检查及免疫组化法检测NF-κB的表达。结果:三组植片存活时间分别为10.17±2.16,19.83±2.25,27.57±1.72d;三组组间比较,差异均有显著性(P<0.01)。A组角膜植片显著水肿、增厚,出现大量的淋巴细胞和单核细胞浸润,角膜基质排列紊乱。B,C两组植片炎性细胞浸润均显著减少,角膜厚度及结构基本正常。NF-κB在A组即对照组中呈高表达(P<0.05),B,C组间比较无显著性差异(P>0.05)。结论:IL-10可有效抑制树突状细胞(dendritic cells,DC)成熟。摄取供体的imDC能显著延长角膜植片的存活时间。IL-10-imDC可抑制NF-κB的表达水平,从而抑制角膜排斥反应的发生。
AIM: To observe the effect of IL-10 modified immature dendritic cells (imDC) for providing theoretical and practical basis for anti-rejection of corneal transplantation. METHODS, Eighteen Wistar rats were used as donors and 36 SD rats were used ad recipients. The recipient was injected by tail with donor IL-10-treated imDC or with untreated imDC. The rats were randomly assigned to three groups, 12 in control group received nopretreatment, 12 in imDC group were injected with imDC (2x 106) 3d before surgery, and 12 in IL-10-imDC group were injected with IL-10-imDC (2x106) 3d before surgery. The survival time of corneal allografts in three groups were observed. The expression of NF - KB was detected by histopathological examination 14d postoperative. RESULTS: The mean survival time was merely 10.17± 2.16d, 19. 83 ± 2. 25d and 27. 57 ± 1. 72d respectively in control, imDC and IL- 10- imDC groups. There was significantly difference among three groups ( P〈 0. 01 ). Corneas in groups B and C had fewer infiltrating cells, corneal thickness almost normal, and stroma arrangement in order, the center of the grafts showed no inflammatory cellular infiltration. NF- KB cells strongly expressed in control group (P〈0.05), and there was no significant difference between groups B and C (P〉0.05). CONCLUSION IL- 10 can restrained DC from mature.Derivatives from donor imDC can markedly prolong the survival time of corneal allografts. NF-KB may play a potential role in the development of corneal graft reiection.
