摘要
目的研究角膜移植后局部淋巴结内树突状细胞数量及趋化因子CCL19含量的变化。方法应用免疫组织化学技术测定大鼠角膜移植后局部淋巴结内趋化因子CCL19含量及树突状细胞数量。结果自体移植组术后局部淋巴结内CCL19含量升高,持续至术后第3天(P<0.05),以后恢复至正常水平(P>0.05)。同种异体移植组,局部淋巴结内CCL19含量在术后1~3d与自体移植组差异无显著性(P>0.05),以后持续上升,至后期显著高于正常组及自体移植组,于第7天达高峰(P<0.05)。树突状细胞数量与CCL19变化趋势一致。结论角膜移植后局部淋巴结内CCL19表达增高,促进树突状细胞的迁移,这可能是同种异体移植排斥反应启动与进展的重要因素。
Objective To investigate alterations in CCL19 content and dendritic cell (DC) numbers in local-draining lymph nodes after penetrating keratoplasty.Methods Rat models of corneal transplantation were established, the content of CCL19 and the number of DC in local-draining lymph nodes were evaluated by immunohistochemical staining.Results The content of CCL19 in local-draining lymph nodes in the isograft group increased within 3 d after surgery ( P < 0.05 ) and then recovered to normal levels ( P > 0.05 ), while the allograft group displayed a continuous trend to increase and migrated into a mass in 7 d (P<0.05). DC numbers showed similar alterations.Conclusion The expression of CCL19 in local-draining lymph nodes is enhanced after allograft, which also accelerates dendritic cell migration and antigen presence. It may be one of the reasons for the induction and promotion of allo-antigen presence when there is a response to rejection.
