摘要
目的研究角膜移植后局部淋巴结内趋化因子CCL19mRNA表达的变化及其对树突状细胞的趋化作用,探讨CCL19在角膜移植中的作用。方法建立大鼠正位值穿透角膜移植模型,RT-PCR法测定正常角膜0d及角膜移植后1d,3d,5d,7d,10d,14d的局部淋巴结内趋化因子CCLI9 mRNA的表达变化,应用Boyden小室进行不同时相淋巴结组织液对体外培养树突状细胞的趋化实验。结果一体移植组术后局部淋巴结内CCL19 mRNA的表达升高,持续至术后3d(P〈0.05),以后恢复至正常水平(P〉0.05)。同种异体移植组,局部淋巴结内CCL19 mRNA的表达术后1~3d与自休移植组差异无统计学意义(P〉0.05),以后持续上升至后期显著高于正常组及自体移植组,于7d达高峰(P〈0.01)。其对体外培养树突状细胞的趋化作用增强(P〈0.05),这种趋化作用可被CCLI9抗体所阻断(P〈0.01)。结论大鼠角膜移植后局部淋巴结内CCL19 mRNA表达增高,促进树突状细胞的迁移可能是同种异体移植排斥反应启动与进展的重要因素。
Objective To investigate the expression of CCL19 mRNA in local draining lymph nodes after penetrating keratoplasty, and their chemotaxls to dendritic cells, and evaluate their effect on corneal allograft survival. Methods Rat models of corneal isograft and corneal allograft were established, The submandibular lymph nodes were harvested before operation and at day 1,3,5,7,10,14 after operation ,The expression of CCL19 mRNA in draining lymph nodes were detected by reverse transcription- polymerase chain reation (RT-PCR), and their chcmotaxis to dendritic cells were examined in Boyden chamber. Resuits the expression of cc119 mRNA in local draining lymph nodes increased within 3d after operation ( P 〈 0,05 ) and there was no statistical difference between isograft and allograft groups ( P 〉 0.05 ) during this perioud. After 3d, the expression of CCL19 mRNA in isograft groups decreased gradully and recovered to normal level ( P 〉 0.05 ), while that of allograft displayed the increasing trend continuously, and reached he peak at 7d, the differences were statistically significant( P 〈 0.01 ), Their homogenate showed enhanced chemotaxis to cultured DC in vitro, which presented the same trend with the expression of CCL19 mRNA, and could be blocked by CCL19 antibody( P 〈 0.01 ), Conclusions The expression of CCL19 mRNA in local draining lymph nodes is ent, anced after allograft, which accelerates dendritic cells migration and antigen presence. It may be one of the reasons to induce and promote allo- antigen presence and rejection response.
