摘要
用碱溶酸沉法、分离蛋白法、盐析法、反胶束法及盐碱两步法这5种方法从苦杏仁中提取蛋白,通过细胞增殖分析实验研究苦杏仁蛋白对人结肠癌细胞生长的影响,结果表明用反胶束法提取的苦杏仁蛋白具有显著抑制人结肠癌细胞生长作用,然而用其他4种方法提取的蛋白没有显示对细胞生长的影响。凝胶过滤层析结果表明,用反胶束法提取的苦杏仁蛋白中含有相对分子量分别约为79、33、24、6ku的4种组分,而用其他4种方法提取的苦杏仁蛋白中含有相对分子量均为72ku的一种组分。SDS-聚丙烯凝胶电泳分析结果表明,用反胶束法提取的蛋白呈现相对分子量为63、51、38、6ku的泳带,与用其他4种方法提取的蛋白呈现泳带存在较大区别。以上结果表明,用反胶束法提取的苦杏仁蛋白中含有对人结肠癌细胞具有毒性作用的特定形式的蛋白质。
Proteins were extracted from apricot kernel by five kinds of method as follow: alkali-solution and acid-isolation, protein isolation, salt fractionation, reverse micelles and salt/alkali two-step method. The effect of the protein on growth of human colon adenocarcinoma cell was studied by cell proliferation assay. The results indicated that the protein extracted from apricot kernel by reverse micelles method inhibited significantly proliferation of the cells, while the proteins from apricot kernel by other four kinds of method were almost inert. Gel filtration chromatography assay of the protein extracted by reverse micelles method showed 79, 33, 24, 6 ku four fractions, and the same assay of the proteins extracted by other four methods exhibited only one fraction of 72 ku. SDS-polyacrylamide gel electrophoresis assay showed four bands of 63, 51, 38, 6 ku on the protein extracted by reverse micelles method lane, which was difference with other four lanes. These results suggest that apricot kernel protein extracted by reverse micelles method contain specific form of protein which could be implicated for cytotoxic activity on Caco-2 cell.
