益智防呆方对Aβ1—40诱导大鼠海马神经元凋亡相关基因表达的影响 被引量:1

摘要 目的观察益智防呆方对Apl-40诱导大鼠海马神经元凋亡相关基因表达的影响。方法大鼠随机分为空白组、模型组、中药小、中、大剂量组及安理申组,双侧侧脑室注射Aβ1-40复制AD大鼠模型。给药4w后提取脑内海马神经元总RNA,RT—PCR测定Caspase-3、Bcl-2、Bax及p53表达。结果与空白组比较,模型组海马神经元Bcl-2表达明显减少妒〈0.01),Caspase-3、Bax、p53表达明显增加俨〈0.01);与模型组比较,益智防呆方能上调海马神经元Bcl-2表达俨〈0.01),下调Caspase-3、Bax、p53表达俨〈0.01)。结论益智防呆方对改善学习记忆障碍的机制可能与上调海马神经元Bcl-2基因表达,下调Caspase-3、Bax、p53基因表达有关。 Objective To investigate effect of Yizhi-Fangdai Decoction on the expression of Aβ1-40-induced apoptosis- related genes in rat hippocampus neural cells. Methods In this study, six animal groups were used, including blank control, model control, groups receiving different concentrations (mild, moderate and high) of herbs and Donepezil group. Experimental AD model was established by intraventricular injection of Aβ1-40. Total RNA ofhippocampal neuron was extracted 4 weeks after treatment administration. Expression of Caspase-3, Bcl-2, Bax and p53 was evaluated using RT-PCR. Results Compared to blank control group, expression of Bcl-2 significantly declined, while Caspase-3, Bax and p53 significantly increased in experimental groups (P〈0.01). Treatment with Yizhi-Fangdai decoction upregulated Bcl-2 level in hippocampal neural cells (P 〈 0.01), and down-regulated Caspase-3, Bax and p53 levels (P〈0.01). Con-clusions Elevated Bcl-2 expression and inhibited Caspase-3, Bax and p53 expression with Yizhi-Fangdai decoction admin- istration could be a possible mechanism that Yizhi-Fangdai decoction improves learning and memory function.
