摘要
目的:观察D-半乳糖衰老大鼠血清睾酮含量及睾丸生精细胞p53基因表达的变化。方法:20只SD大鼠随机分为正常组、衰老模型组,每组均为10只。采用D-半乳糖连续腹腔注射制作亚急性衰老的大鼠模型。ELISA方法检测血清睾酮含量,SP免疫组化法检测睾丸生精细胞p53基因的表达。结果:衰老大鼠血清睾酮含量明显下降(与正常大鼠相比:P<0.01);衰老大鼠睾丸p53阳性细胞率明显高于正常大鼠(P<0.01)。结论:衰老大鼠睾丸生精细胞p53基因表达上调,睾丸功能减退。
Objective: To observe the changes of testosterone content in serum and p53 gene expression of spermatogenic cells in testis of D-gal aging rats. Methods:20 SD rats were applied in this study and divided into 2 groups at random: normal group, aging model group, and each group had 10 rats. The sub-acutely aging model rats were made by injecting D-gal into abdominal cavity continually. ELISA was used to detect the testosterone content in serum, SP immunohistochemistry to observe the p53 expression of spermatogenic cells.Results: The testosterone content of aging rats decreased significantly compared with normal rats (P 〈0.01); the p53 positive cell rate of aging rats were obviously higher than normal rats (P〈0.01). Conclusions: The p53 gene expression of spermatogenic cells in testis of aging rats increased, and the function of testis decline.
