摘要
目的:探讨骨性关节炎(osteoarthritis,OA)患者成纤维样滑膜细胞(fibroblast-like synoviocytes,FLS)Notch信号途径相关分子表达是否存在异常。方法:体外分离培养滑膜FLS,采用LPS或LPS联合Notch信号阻断剂DAPT处理3~5代FLS,应用RT-PCR检测滑膜细胞Notch受体及下游靶基因Hes-1 mRNA的表达。采用蛋白质印迹法检测Hes-1蛋白质表达水平。结果:与正常人相比,OA患者成纤维样滑膜细胞Notch受体1、2、3以及Hes-1 mRNA表达水平明显增高(P<0.05);LPS处理可显著上调OA患者FLS的Notch受体1、2、3(Notch4表达量较低)及Hes-1表达。DAPT处理可下调OA患者FLS的Hes-1蛋白质表达水平。结论:Notch信号途径在OA患者FLS中呈现明显的活化状态,其在OA关节局部炎性机制中可能发挥重要作用。
Objective: To investigate the expression of Notch signaling molecules in fibroblast-like synoviocytes(FLS) from patients with osteoarthritis(OA).Methods: OA FLSs were isolated from synovial tissues and treated with LPS or LPS + DAPT between the passages of 3 and 5.RT-PCR was used to detect the expression of mRNA of the Notch receptors and Notch target gene Hes-1.In addition,the protein level of Hes-1 was detected by Western blot.Results: Compared with normal FLSs,the mRNA expression of Notch receptors 1,2,3 and Hes-1 was markedly increased in OA FLSs(P&lt;0.05).LPS treatment significantly up-regulated the expression of Notch receptors1,2,3(Notch 4 level was low) and Hes-1.However,DAPT,a Notch signaling inhibitor,could down-regulated the protein level of Hes-1.Conclusion: Notch signaling was found to be activated in OA FLS,which may be involved in the pathophysiology of the inflammatory in OA joints.
