摘要
目的探讨Notch信号通路中的靶基因Hes-1和IFN-α、IFN-β、IFN-γ三种干扰素与丙型肝炎病毒(HCV)感染的关系。方法选取2012年11月—2014年10月在我院就诊的125例HCV感染患者,根据是否转归为肝硬化分为HCV感染组和HCV肝硬化组,同期选取100例健康人为对照组。采集HCV肝硬化组、HCV感染组和对照组晨起空腹静脉血3 ml,分离出单个核细胞,提取RNA,将其反转录为c DNA,应用实时定量RT-PCR技术检测Hes-1和IFN-α、IFN-β、IFN-γ。结果 3组Hes-1和IFN-α、IFN-β、IFN-γ水平之间比较,差异具有统计学意义(P<0.05)。结论 Notch信号通路中的靶基因Hes-1和IFN-α、IFN-β、IFN-γ在HCV慢性感染时明显升高,且在HCV感染导致的肝硬化中,Hes-1和IFN-α、IFN-β、IFN-γ的水平更高。
Objective To investigate the relationships between Hes-1 target gene of Notch signaling pathways and interferon-α( IFN-α),IFN-β and IFN-γ and hepatitis C virus( HCV) infection. Methods A total of 125 HCV inpatients admitted during November 2012 and October 2014 were divided into hepatic cirrhosis group( HC group,n = 32)and HCV infection group( HCV infection group,n = 93),while 100 healthy people at the same period were selected as control group. Samples of 3 ml venous blood with empty stomach in the morning were obtained from HC group,HCV infection group and control group,and mononuclear cells were isolated,RNA was extracted,and then obtained the reverse transcription of c DNA was performed. The levels of Hes-1,IFN-α,IFN-β and IFN-γ were detected using real-time quantitative RT-PCR( reverse transcription polymerase chain reaction) method. Results The differences in levels of Hes-1,IFN-α,IFN-β and IFN-γ in the three groups were statistically significant( P〈0. 05). Conclusion Hes-1 target gene of Notch signaling pathways and Hes-1,IFN-α,IFN-β and IFN-γ levels are significantly increased in HCV patients with chronic infection,but the levels are even more increased in hepatic cirrhosis induced by HCV infection.
