摘要
目的:观察健脑通胶囊对AD大鼠单胺氧化酶(MAO)的影响,从免疫组化方面探讨其作用机制。方法:从84只SD大鼠中随机选出12只(雌雄各半)为正常对照组,剩余72只进行AD模型复制,造模成功后死亡3只,剩余69只随机分成5组:AD模型组(12只)、脑复康组(12只)、健脑通胶囊小剂量组(15只)、健脑通胶囊中剂量组(15只)、健脑通胶囊大剂量组(15只);健脑通胶囊大、中、小剂量组分别予健脑通胶囊0.94g.kg-1·d-1、0.47g.kg-1·d-1、0.235g.kg-1·d-1灌胃,西药组给予脑复康片混悬液灌胃,剂量为50mg/mL,正常对照组予以等量生理盐水灌胃,每天上午给药1次,连续30d。结果:健脑通胶囊各组与脑复康组脑组织中单胺氧化酶(MAO)含量均高于模型组(P<0.05),且健脑通胶囊大、中剂量组优于小剂量组,差异有统计学意义(P<0.05)。结论:健脑通胶囊能减少AD模型大鼠单胺氧化酶(MAO)含量,延缓老年痴呆的发展,从而有效保护脑功能。
Objective:To observe the effect of Jiannaotong capsules on monoamine oxidase(MAO) level in rats with Alzheimer~ dis- ease(AD) and to investigate its action mechanism from the aspect of immunohistochemistry. Methods:Twelve rats (female -to -male ratio = 1 : 1 ) were randomly selected from 84 AD rats as normal control group, and the other 72 rats were treated to establish an AD model. Three rats died while the model was induced. Sixty- nine surviving rats were randomly divided into AD model group( n = 12 ) , piracetam group( n = 12 ) , low - dose Jiannaotong capsule group ( n = 15 ) , middle - dose Jiannaotong capsule group ( n = 15 ) , and high - dose Jiannaotong capsule group (n = 15 ). The high - , middle - , and low - dose Jiannaotong capsule groups received Jiannaotong capsules (0. 94g·kg^-1·d^-1 ,0.47 g/kg· d, and 0. 235 g/kg· d, respectively) by garage, the piracetam group received suspension of piracetam tablets (50 mg/ml) by gavage, and the normal control group received an equal volume of saline by garage. Medication was given once in the morning every day for 30 consecutive days. Results:The Jiannaotong capsule groups and piracetam group had significantly higher MAO levels in brain tissue than the model group( P 〈 0. 05 ) , and the high - and middle - dose Jiannaotong capsule groups were signifi- cantly superior to the low - dose Jiannaotong capsule group in this regard ( P 〈 0.05 ). Conclusion : Jiannaotong capsules can decrease MAO level in brain tissue in AD rats, thus delaying the progression of AD and protecting brain function.
