摘要
目的 探讨SCID鼠与免疫学相关的外周血指标变化及个体“渗漏”问题。方法 采用血细胞全自动分析仪检测SCID、NC、BALB/c鼠 14项外周血指标 ,3H TdR掺入测T、B淋巴细胞转化功能和细胞毒活性。结果 SCID鼠与NC、BALB/c鼠相比 ,WBC、淋巴细胞分类 (Ly)的比例显著减少 ;中性粒细胞 (GR)和红细胞体积分布宽度 (RDW )显著增加。随着鼠龄的增长 ,WBC和Ly增加 ,GR和RDW减少 ,其中变化最显著的个体 (WBC≥ x±2SD)称之为“渗漏” ,在鼠龄 6~ 8周时和 17~ 19周时的渗漏率分别为 5 .5 %和 2 6 .7%。此外 ,SCID鼠的T、B淋巴细胞转化和CTL细胞功能与BALB/c鼠相比显著低下。结论 SCID鼠T、B淋巴细胞联合缺陷 ,可在免疫生物学领域中广泛应用 ,但要及时剔除个体“渗漏”
Objective To probe into peripheral blood index variety and individual “leakage” of mice which is connected to immunology.Methods Fourteen peripheral blood indexes were determined with the blood cell automatic analysing machinery.T、B lympocyte transformation function and cytotoxicity activity were determined by mixing HTdR.Results Compared with ones of NC,BALB/c,the proportion of white blood cell (WBC) and lymphocyte (Ly) of SCID mice decreased obviously;but,neutrophil granulocyte (GR) and red blood cell distribution width (RDW) of SCID mice increased obviously.As the mice growing,WBC,Ly increase and GR、RDW decreased.The individual which changed mostly (WBC≥±2SD) was called as “Leakage”.The “Leakage”rate in the 6~8 weeks mice and 17~19 weeks mice were 5.5% and 26.7%;besides,compared with ones of BALB/c mice,the T、B lymphocyte transformation and CTL cell function in SCID mice decreased obviously.Conclusion SCID mice lacks T,B lymphocyte and can be used broadly in realm of immunobiology.But,the individual “Leakage” mice must get rid of in time.
