淫羊藿苷对前列腺癌原位移植瘤SCID小鼠脂肪酸合成酶和LNCaP细胞生物学行为的影响 被引量:3

摘要 目的观察淫羊藿苷(Icariin,ICA)对前列腺癌原位移植瘤SCID小鼠前列腺肿瘤组织中脂肪酸合成酶(Fatty acid synthase,FAS)mRNA和LNCa P细胞生物学行为的影响。方法 6周龄雄性SCID小鼠30只,从左、右前列腺背外侧叶包膜内注射复苏的人LNCa P前列腺癌细胞株(2×10~7个/mL)悬液各25μL,建立SCID小鼠前列腺癌原位移植瘤模型,2周后随机均分为前列腺癌组(CaP组)、淫羊藿苷高剂量组(80 mg/kg,高剂量组)和淫羊藿苷低剂量组(40 mg/kg,低剂量组),另取健康6周龄雄性SCID小鼠10只设为空白对照组(CON组)。于治疗结束后测量各组SCID小鼠前列腺原位移植瘤瘤体积、瘤重量,计算抑瘤率;采用免疫组化法检测FAS在前列腺肿瘤组织中的阳性蛋白表达,RT-PCR检测FAS mRNA表达,流式细胞学(Flow cytometry,FCM)检测LNCaP细胞周期,以分析ICA对LNCa P细胞生物学行为的影响。结果 ICA可明显抑制前列腺癌原位移植瘤的生长,高剂量组和低剂量组瘤体积[(0.45±0.04)、(0.47±0.06)cm^3]和瘤重量[(0.69±0.07)、(0.71±0.09)g]明显低于CaP组,FCM检测显示,抑瘤率分别为39.47%和37.28%,且两组S期细胞比值升高(50.34%±1.53%、47.56%±2.67%)、G_0/G_1期细胞比例降低(25.40%±1.84%、26.51%±2.21%),与CaP组比较差异有统计学意义(P<0.05),但两组组内比较差异无统计学意义(P>0.05);FAS蛋白(21.04%±2.73%、22.49%±1.94%)及FAS mRNA[(13.87±1.12)、(12.73±2.07)/GAPDH]在高剂量组和低剂量组前列腺肿瘤组织中均低表达,与Ca P组比较差异有统计学意义(P<0.05),但两组组内比较差异无统计学意义(P>0.05)。结论 ICA可能通过抑制FAS活性、改变细胞周期分布(阻滞于S期)、诱导肿瘤细胞凋亡和生长抑制,有效抑制前列腺癌LNCa P细胞增殖,并降低细胞侵袭能力。 Objective To observe the effects of icariin( icariin,ICA) on biological behaviors of LNCaP cells and mRNA of fatty acid synthase( FAS) in prostate tissue of SCID mice with prostate cancer orthotopic transplantation tumor. Methods Totally thirty 6-week old male SCID mice were injected revived human LNCa P prostate cancer cell lines( 2 × 10^7/m L) suspension 25 μL from the left and right dorsolateral prostate lobe capsule to make orthotopic transplantation tumor model,and after 2 weeks they were randomly divided into CaP group( prostate cancer group),icariin high-dose group( 80 mg/kg) and icariin low-dose group( 40 mg/kg); another ten 6-week old healthy male SCID mice were chosen as blank control group( CON group). At the end of the treatment,tumor volume and weight of prostate orthotopic transplantation tumor of SCID mice were measured,and the inhibition rate was calculated. Immunohistochemistry was used to detect the expression of FAS in prostate cancer protein; RT-PCR was used to detect the expression of FAS fabric mRNA;flow cytometry( FCM) was used to detect the cycle and apoptosis rate of LNCaP cell in order to analyze the influences of ICA on the biological behavior of LNCaP cells. Results ICA could significantly inhibit the growth of orthotopic transplantation tumor of prostate cancer; tumor volumes [( 0. 45 ± 0. 04) and( 0. 47 ± 0. 06) cm^3] and tumor mass [( 0. 69 ±0. 07) and( 0. 71 ± 0. 09) g] of high-dose group and low-dose group were significantly lower than those of CaP group.The FCM detection showed that the inhibition rate was 39. 47% and 37. 28%,respectively; in the two icariin groups,the cells ratio of S phase was higher( 50. 34% ± 1. 53% and 47. 56% ± 2. 67%),and the proportion of cells in G0/G1 phase decreased( 25. 40% ± 1. 84% and 26. 51% ± 2. 21%),there being statistical difference compared with CaP group( P〈0. 05); however,there was no significant difference between the two groups( P〉0. 05).
