摘要
目的:研究玉米肽(CPs)体内外的抗肿瘤作用。方法:选用人肝癌HepG2细胞进行体外实验,经CPs处理HepG2细胞后,借助透射电镜观察细胞形态学变化;在激光共聚焦显微镜下观察细胞内Ca2+浓度的变化;选用BABL/c小鼠进行体内实验,接种小鼠肝癌系H22细胞,考察CPs对H22荷瘤小鼠瘤质量、免疫器官指数、血清SOD活性及肝脏MDA含量的影响。结果:透射电镜观察显示,CPs作用48h后,HepG2细胞形态变化明显;同时细胞内的Ca2+浓度增加;CPs中、高剂量组能抑制H22荷瘤小鼠体内的肿瘤生长(P<0.05;P<0.01),CPs各剂量组均能增加小鼠胸腺指数,并提高小鼠脾脏指数、血清中SOD活性(P<0.01),降低其肝脏内MDA含量(P<0.01)。结论:CPs在体外具有诱导细胞凋亡作用,体内有抑制肿瘤增殖的作用。
To explore the antitumor activity of com peptides (CPs) in vitro and in vivo, the morphology of HepG2 cells and the change of Ca2+ concentration caused by CPs were observed by transmission electron microscopy and laser confocal microscope, respectively. Meanwhile, H22 tumor-bearing mice were administered with CPs at different dosages by intraperitoneal injection for 10 d. The anti-tumor activity of CPs was evaluated by tumor weight, thymus index and spleen index, SOD activity and MDA content in tumor-bearing mice. The results showed that apoptosis characteristics such as chromatin condensation, accumulation to the nuclear edge and cytoplasmic condensation, and increased intracellular Ca2; concentration were obvious in HepG2 cells after CP treatments. Moreover, CPs could not only significantly suppress the growth of H22 tumor in mice (P 〈 0.05; P 〈 0.01) and increase spleen index (P 〈 0.01), but also improve SOD activity in the serum (P 〈 0.01), and MDA content (P 〈 0.01) in the liver of H22-bearing mice. Therefore, corn peptides can be a safe and effective anticancer, bioactive agent or functional food.
