摘要
应用胃肠道蛋白酶水解菜籽分离蛋白,并通过膜分离技术将水解物分离成4种不同分子量组分(<1 kDa、1~3 kDa、3~5 kDa和5~10 kDa);研究了水解物和不同组分对HepG2、He-La和MCF-7三种肿瘤细胞的体外增殖抑制作用及其氨基酸组成.结果表明:水解物及各组分对HepG2体外增殖具有较明显的抑制作用,而对HeLa和MCF-7体外增殖的抑制效果不明显;<1 kDa的组分对HepG2的抑制率最强(36.4%),其氨基酸组成中以谷氨酸、亮氨酸、天冬氨酸含量最高.研究认为,菜籽蛋白胃肠道蛋白酶水解物及其<1 kDa的膜分离组分可能作为功能性成分用于抗肿瘤相关的功能食品和保健品的开发.
Rapeseed protein isolate was hydrolyzed by pepsin & pancreatin and then separated into four different fractions (〈1 kDa, 1-3 kDa, 3-5 kDa, and 5-10 kDa) by membrane ultra- filtration. Growth inhibitions to HepG2, HeLa and MCF - 7 in vitro of rapeseed protein hydrolysate and membrane ultrafiltration fractions were employed to evaluate their activities. The results show rapeseed protein hydrolysate and membrane ultrafiltration fractions inhibit HepG2 growth in vitro significantly but don't have significant growth inhibition effects against HeLa and MCF- 7 in vitro. Fraction %1 kDa possesses the highest inhibition rate against HepG2 (36.4%) and the contents of Glu, Leu and Asp are the top 3 in its amino acid composition. The results suggest that rapeseed protein hydrolysate and fractions % 1 kDa can be used to develop the functional foods and health products that can inhibit tumor growing.
