游泳对大鼠左心室心肌超微结构的影响

摘要 目的探讨不同运动量游泳运动对大鼠左心室心肌细胞超微结构的影响,以期为运动对心肌的生理和病理变化研究提供参考资料。方法 SD大鼠分为不同运动量运动组和力竭运动组,前者进行12周的低、中和高运动量游泳运动,后者进行1次力竭游泳运动。取左心室心肌组织进行常规透射电镜样品制备,透射电镜观察左心室心肌细胞超微结构的变化。结果低运动量运动对大鼠左心室心肌细胞结构影响不大;中运动量运动提高心肌肌节长度,线粒体数量和体积增加,嵴变得较密集;高运动量运动对心肌细胞产生轻微破坏,肌节变短,核膜内陷,核质凝缩,线粒体嵴疏松模糊。力竭运动后即刻,对心肌细胞的细胞核和线粒体影响较大,肌节长度缩短。力竭后24 h,心肌细胞出现细胞核和线粒体损伤减轻,肌节长度增加的变化,但肌纤维排列严重紊乱。结论高运动量和力竭运动可以导致左心室心肌细胞超微结构破坏。 【Objective】 To explore the effects of different loading swimming exercises on ultrastructure of myocardial cell of ventriculus sinister in rat,so as to provide some information for the changes of physiology and pathology by the exercise-induced injury and recuperation of cardiac muscle.【Methods】 SD rats were randomly divided into two groups: different loading exercise group and exhaustive exercise group.In the different loading exercise group,a 12-week low,moderate and high loading swimming model was carried out in rats.In the exhaustive exercise group,a single exhaustive swimming was carried out in rats.After exercises,rat's ventriculus sinisters were cut off,and treated with routine procedures of transmission electron microscopy before observing the ultrastructure of myocardial cell under transmission electron microscope.【Results】 For low loading swimming exercise rats,the myocardial cell of ventriculus sinister showed intact clear ultrastructure.For moderate loading swimming exercise rats,the length of sarcomere increased,the quantities and volumes of mitochondria increased,and their cristae were densely arranged.For high loading swimming exercise rats,the ultrastructure of myocardial cell was lightly destroyed,the length of sarcomere decreased,nuclear envelope invaginated,chromatin condensed,and the mitochondrion cristae became unclear.At the end of exhaustive swimming exercise,the nucleus and mitochondrion were heavily changed,and the length of sarcomere decreased.At 24 hours after exhaustive exercise,the ultrastructure of nucleus and mitochondria showed attenuated injury,and the length of sarcomere increased,but the myocardial fibrils were arranged in heavy disorder.【Conclusion】 High loading and exhaustive exercises could destroy the structure of myocardial cell of ventriculus sinister.
