摘要
目的:观察大鼠力竭性游泳后窦房结病理组织学和缺血缺氧性变化。方法:实验于2004-09/12在扬州大学医学院解剖研究室完成。①选用8周龄的SD大鼠32只,雌雄各半。采用300L塑料水箱作为大鼠游泳槽,水深60cm,约为大鼠身长2倍,水温控制在(34±1)℃。按随机抽签法将大鼠分为4组:安静对照组(不运动),中等强度训练组(无负重游泳,120min/d,共7d),一次力竭训练组(尾部负荷体质量的2%,进行1次力竭游泳)和1周力竭训练组(尾部负荷体质量的2%,每天1次力竭游泳,共7d)。力竭标准:大鼠连续3次沉入水底,每次超过10s,不能自主浮上水面。②运动后即刻摘取窦房结,经固定、切片后分别进行苏木精染色、Masson’s染色和Nagar-Olsen染色。③用图像分析仪检测窦房结内P细胞、T细胞、心房肌细胞、胶原纤维胞浆吸光度(A值)。④组间计量资料差异比较采用t检验。结果:大鼠32只均进入结果分析。①苏木精-伊红和Masson’s染色显示,对照组窦房结内各细胞形态正常;中等强度训练后窦房结内部各细胞的形态结构无明显改变;1次力竭训练后窦房结内P细胞、T细胞与心房肌细胞病理改变明显,而胶原纤维的改变不明显。1周力竭训练后窦房结内P细胞、T细胞与心房肌细胞病理改变减轻,但胶原纤维的染色强度增加。中等强度训练后窦房结内各细胞胞浆A值与对照组相近(P>0.05)。1次力竭训练组窦房结内P细胞、T细胞和心房肌细胞胞浆A值均明显低于对照组和中等强度训练组(P<0.05~0.01)。1周力竭训练组窦房结内各细胞胞浆A值明显低于对照组和中等强度训练组(P<0.05),T细胞和心房肌细胞胞浆A值明显高于1次力竭训练组(P<0.05)。1周力竭训练组窦房结内胶原纤维胞浆A值明显高于其他3组(P<0.05)。②Nagar-Olsen染色(反映窦房结组织缺血缺氧程度)显示,对照组与中等强度训练组的窦房结内部细胞无缺血缺氧改变;
AIM: To observe the changes in pathohistology and ischemic hypoxia of sinoatrial node in rats after exhaustive swimming.
METHODS: The experiment was accomplished in the Department of Anatomy of Medical College of Yangzhou University from September to December 2004. ①Thirty-two male and female SD rats, aged 8 months, were selected. Plastic water tank (300 L) was taken as the swimming, pool of rats with the depth of 60 cm, which was two times of rat's body length, and the temperature was (34±1) ℃. Rats were randomly divided into 4 groups: control group (without exercise), medium intensity exercise group (swimming without weight loading, 120 minutes a day for 7 days), once exhaustive swimming group, (once exhaustive swimming with tail-loaded, which was 20% of the body mass) and one-week exhaustive swimming group (exhaustive swimming with tail-loaded, which was 20% of the body mass once a day for 7 continuous days). Exhaustive standard: rats sarik into the water for 3 times with each time over 10 seconds, and couldn't float.② The sinoatrial nodes were removed immediately after the exercise, and then respectively dealt with hematoxylin stain, Masson's stain and Nagar-Olsen stain after being fixed and cut into sections. ③The P cells and T cells inside the sinoatrial node, atrial muscle cells and absorptance (A) value of cytoplasm in collagen fibers were detected with image analyzer. ④ Differences of measurement data among groups were compared with t test.
RESULTS: A total of 32 rats were involved in the analysis of results.① Hematoxylin-eosin stain and Masson's stain showed that all the cellular shape in sinoatrial nodes of the control group were normal, and those in the medium intensity exercise group were not significantly changed. The pathological changes of P cells, T cells and atrial muscle cells in sinoatrial nodes of once exhaustive swimming group were remarkable, whereas the changes of collagen fibers were not marked.
