万寿菊茎叶中2种黄酮类化合物的体外抗肿瘤活性 被引量:12

摘要 目的:研究万寿菊茎叶中两种黄酮类化合物4'-甲氧基-泽兰素-3-O-β-D-葡萄糖苷(化合物Ⅰ)和山柰酚-3,7-O-α-L-双鼠李糖苷(化合物Ⅱ)的抗肿瘤活性。方法:人胃癌细胞SGC7901与人肝癌细胞SMMC7721经20,40,80,120,160μmol.L-1的两种化合物作用24,48,72 h后,使用MTT法测定抑制率,并以5-氟脲嘧啶作为阳性对照。使用吖啶橙/溴化乙锭双荧光染色法观察浓度为160μmol.L-1的两种化合物作用于细胞密度为1×106个/mL的2种肿瘤细胞48 h细胞形态学变化及细胞凋亡情况。结果:不同浓度的两种黄酮类化合物均可抑制人胃癌细胞SGC7901和人肝癌细胞SMMC7721的增殖,且呈现浓度与时间依赖性。化合物Ⅰ作用这2种癌细胞48 h的半数抑制浓度(IC50)分别为111.7,330.4 mg.L-1。化合物Ⅱ作用这两种癌细胞48 h的IC50分别为683.8,464.7 mg.L-1。2种化合物作用肿瘤细胞后细胞形态发生改变,并有部分细胞发生凋亡。结论:万寿菊茎叶中分离的2种黄酮类化合物具有体外抗肝癌和胃癌活性。 Objective: To study the anti-cancer activity of two flavonoids 4'-methoxy-eupatolitin-3-O-βD-glucoside(compoundⅠ) and kaempfrol-3,7-O-α-L-di-rhamnoside(compound Ⅱ) in the leaf and stem of Tagetes erecta.Method: SGC7901 and SMMC7721 cells were treated by 20,40,80,120,160 μmol.L-1 two flavonoids for 24,48,72 h,MTT assay was used to determine the inhibition rates,and 5-FU was used as positive control.AO / EB double fluorescent staining was used to observe the changes of cell morphology and the apoptosis of tumor cells at the concentration of 1 × 106/ mL after treated with 160 μmol.L-1 of two flavonoids for 48 h.Result: The different concentrations of two flavonoids could both inhibit the proliferation of SGC7901 and SMMC7721 in the time-and dose-dependent manner.The IC50 of compoundⅠ was 111.7 mg.L-1 and 330.4 mg.L-1,respectively.The IC50 of compound Ⅱ was 683.8 mg.L-1 and 464.7 mg.L-1,respectively.The morphology of two tumor cancer cells was changed after treated by two compounds,and part of them emerged apoptosis.Conclusion: Two flavonoids separated from the leaf and stem of T.erecta have the activity of anti-liver cancer and anti-gastric cancer in vitro.
